摘 要:目的探讨卵巢癌组织中微小RNA-338-3p(miR-338-3p)及结肠癌转移相关基因1(MACC1)的表达变化及其临床意义。方法选取行手术治疗的卵巢癌患者91例,同期选择子宫良性疾病患者30例作对照。术中取卵巢癌组织及对照患者卵巢正常组织,用qRT-PCR法检测以上组织中miR-338-3p、MACC1 mRNA表达,分析二者表达与卵巢癌患者临床病理参数的关系,用Pearson线性相关分析miR-338-3p与MACC1 mRNA表达的相关性;术后即对卵巢癌患者进行门诊随访,分析miR-338-3p、MACC1 mRNA表达与卵巢癌患者预后的关系。结果卵巢癌组织中miR-338-3p相对表达量低于卵巢正常组织(t=229.316,P=0.000),MACC1 mRNA相对表达量高于卵巢正常组织(t=243.188,P=0.000)。卵巢癌组织中miR-338-3p与MACC1 mRNA表达呈负相关(r=-0.621,P=0.000)。卵巢癌组织中miR-338-3p、MACC1 mRNA相对表达量与肿瘤分化程度、FIGO分期及淋巴结转移有关(P均<0.05)。miR-338-3p高表达患者5年总体生存率高于miR-338-3p低表达患者(P均<0.05),MACC1高表达患者5年总体生存率低于MACC1低表达患者(P均<0.05)。结论卵巢癌组织中miR-338-3p呈低表达,而MACC1呈高表达;二者可能参与肿瘤的发生发展,并影响患者预后。Objective To study the expression changes and clinical significance of microRNA(miR)-338-3p and metastasis-associated in colon cancer-1(MACC1)in ovarian cancer tissues.Methods Ninety-one patients with ovarian cancer underwent surgical treatment and 30 patients with benign uterine disease were selected as controls.The expression levels of miR-338-3p and MACC1 in the ovarian cancer tissues and normal ovarian tissues were detected by qRT-PCR.The relationship between the expression of miR-338-3p and MACC1 and clinicopathological characteristics was analyzed.Pearson linear correlation was used to analyze the correlation between miR-338-3p and MACC1 mRNA expression.After the operation,the patients with ovarian cancer were followed up in the clinic to analyze the relationship between the expression of miR-338-3p,MACC1 mRNA and the prognosis of patients with ovarian cancer.Results The relative expression of miR-338-3p in the ovarian cancer tissues was lower than the normal ovarian tissues(t=229.316,P=0.000),and the relative expression of MACC1 mRNA was higher than that of the normal ovarian tissues(t=243.188,P=0.000).Pearson linear correlation analysis showed that miR-338-3p was negatively correlated with MACC1 mRNA expression in the ovarian cancer tissues(r=-0.621,P=0.000).The relative expression of miR-338-3p and MACC1 in the ovarian cancer tissues was related to the degree of tumor differentiation,FIGO stage and lymph node metastasis(all P<0.05).The 5-year overall survival rate of patients with ovarian cancer with high miR-338-3p expression was significantly higher than that of patients with low miR-338-3p expression(P<0.05).The 5-year overall survival rate of patients with high MACC1 expression was significantly lower than that of patients with low MACC1 expression(P<0.05).Conclusion MiR-338-3p is low expressed in the ovarian cancer tissues,while MACC1 is highly expressed,and both of them may be involved in the occurrence and development of tumor and affect the prognosis of patients.
