摘  要:通过单因子试验和多因子的正交试验对解淀粉芽孢杆菌亚种LE-3液体摇瓶发酵产胞外果聚糖的培养条件(碳源、氮源、接种量、发酵时间)进行了优化,并提取纯化果聚糖,进一步测定了该果聚糖螯合Fe2+的能力.结果表明:解淀粉芽孢杆菌亚种LE-3种子菌液培养24 h后按照10%(v/v)接种发酵培养基(蔗糖60 g/L、酵母膏2.5 g/L、KH2PO42.5 g/L、K2HPO42.5 g/L、NaCl 1.0 g/L、MgSO4·7H2O 0.2 g/L、FeSO4·7H2O 0.001 g/L,初始pH 7.0),37℃培养36 h,胞外果聚糖产量达到最高值18.36 g/L,约是未优化时的3.0倍.The medium and fermentation conditions, including carbon source, nitrogen source, size of the inoculum and fermentation time, to produce fructan by Bacillus amyloliquefaciens subsp. amyloliquefaciens LE-3 were optimized by single factor and orthogonal array experiments. At the same time the fructan produced by strain LE-3 was abstracted and purified, and then its ability to chelate Fe2+ was measured. Results showed that after culture at 37 ℃ for 36 h in a medium consisting of sucrose 60 g/L,yeast extract 2.5 g/L, KH2PO4 2.5 g/L, K2HPO4 2.5 g/L, NaCl 1.0 g/L, MgSO4· 7H2O 0.2 g/L and FeSO4· 7H2O 0.001 g/L, a maximum fructan production of 18.36 g/L was achieved, which was 3.0 times higher than that before optimization.
