摘  要:目的:明确固有免疫受体NOD1对人早孕期滋养细胞侵袭功能的调控及对侵袭相关因子分泌的影响。方法:采用免疫细胞化学法鉴定原代滋养细胞NOD1的表达,用transwell侵袭实验检测激活NOD1后滋养细胞侵袭功能的改变,ELISA检测配体刺激后滋养细胞MMP2和MMP9的分泌情况。结果:免疫细胞化学结果显示滋养细胞分离鉴定成功,且原代滋养细胞可以表达固有免疫受体NOD1。使用NOD1的特异性配体及非特异性配体,发现激活NOD1可以抑制滋养细胞的侵袭,且非特异性配体LPS可以下调侵袭相关金属基质蛋白酶分子MMP2和MMP9的分泌,特异性配体i E-DAP仅下调MMP9的分泌而对MMP2的分泌无影响。结论:固有免疫模式识别受体NOD1可以在早孕期滋养细胞表达,可调控滋养细胞的侵袭功能,其激活会导致侵袭相关分子MMP2和MMP9的分泌下降。Objective: To identify the effect of innate immunity receptor NOD1 on the invasion of human early pregnancy trophoblast cells. Methods: The expression of NOD1 was identified by immunocytochemistry. Transwell matrigel invasion assay was used to analyze the ability of invasiveness after NOD1 activation. We also used ELISA assay to test the secretion of MMP2 and MMP9 after NOD1 activation. Results: Immunocytochemistry result showed the separation of human trophoblast cells was successful. NOD1 had been expressed by primary trophoblast cells. Activation of NOD1 inhibited the invasion of trophoblasts. LPS down-regulated the secretion of both MMP2 and MMP9 while i E-DAP only down-regulated the secretion of MMP9. Conclusion: Innate immune pattern recognition receptor NOD1 was expressed by trophoblast cells in early pregnancy, which could regulate the invasion of trophoblasts.
