摘 要:目的探讨阿尔茨海默病大鼠海马组织中基质金属蛋白酶-9(MMP-9)和尿型纤溶酶原激活剂(uPA)的表达及黄芪颗粒的干预作用。方法建立阿尔茨海默病大鼠的动物模型,大鼠随机归为4组:对照组、模型组、黄芪组及西药组。西药组和黄芪组分别口服黄芪颗粒和吡拉西坦,对照组与模型组仅口服等剂量的生理盐水,治疗时间为30天。水迷宫实验评估每组鼠的逃避潜伏期和穿台次数,免疫印迹实验测定各组鼠海马组织中MMP-9和uPA的含量。结果与对照组相比,模型组大鼠的逃避潜伏期[(47.24±3.12)s]延长,穿台次数(8.16±2.68)减少(均P<0.05),MMP-9(0.801±0.056)和uPA(0.699±0.047)含量的相对值明显上升(均P<0.05)。与模型组相比,口服黄芪颗粒和吡拉西坦的大鼠逃避潜伏期[(39.67±2.11)和(40.11±2.12)s]缩短,穿台次数(10.91±2.08、11.09±2.34)增加(均P<0.05),MMP-9(0.342±0.029、0.367±0.037)和uPA(0.251±0.031、0.246±0.028)含量的相对值均显著下降(均P<0.05)。结论黄芪颗粒可通过降低阿尔茨海默病大鼠MMP-9和uPA的含量来改善其学习与记忆功能。Objective To explore the expression of matrix metalloproteinase-9(MMP-9)and urinary plasminogen activator(uPA)in hippocampus of rats with Alzheimer’s disease and the intervention effect of astragalus granules.Methods An animal model of Alzheimer’s disease in rats was established,and the animals were separated into four groups:control group,model group,astragalus group and western medicine group.Astragalus group and western medicine group were given Astragalus granules and pyroacetamide respectively,while the control group and model group only took the same amount of sterile saline orally,and the treatment time was 30 days.Morris water maze was used to measure the positioning navigation and space exploration abilities of each group of rats.The contents of MMP-9 and uPA in hippocampus of rats in each group were estimated by immunoblotting.Results Compared with the control group,the escape latency[(47.24±3.12)s]in model group extended,frequency of crossing(8.16±2.68)decreased(both P<0.05),and the relative contents of MMP-9(0.801±0.056)and uPA(0.699±0.047)significantly increased(both P<0.05).Compared with the model group,the escape latency in astragalus group and western medicine group[(39.67±2.11)and(40.11±2.12)s]shortened,frequency of crossing(10.91±2.08,11.09±2.34)increased(all P<0.05),and the relative contents of MMP-9(0.342±0.029,0.367±0.037)and uPA(0.251±0.031,0.246±0.028)significantly decreased(all P<0.05).Conclusion Astragalus granules can improve learning and memory function by reducing the contents of MMP-9 and uPA in rats with Alzheimer’s disease.
