摘  要:目的观察偏治颗粒对硝酸甘油诱导偏头痛大鼠模型的防治作用及机制。方法将60只SD大鼠按照随机数字表法分为6组,即空白组、模型组、偏治颗粒低剂量组(5.4 g生药/kg)、偏治颗粒中剂量组(10.8 g生药/kg)、偏治颗粒高剂量组(20.16 g生药/kg)、对照组(琥珀酸舒马普坦片18 mg/kg),每组10只。偏治颗粒低、中、高剂量组及对照组大鼠均预防给药7 d,每日1次灌胃,给药容积为10 mL/kg,空白组、模型组均予等容积蒸馏水。末次灌胃1 h后,除空白组外,其余5组大鼠均皮下注射硝酸甘油10 mg/kg,制备偏头痛大鼠模型。观察造模后各组大鼠耳红出现及消失时间,比较各组大鼠三叉神经脊束核神经元降钙素基因相关肽1型(CGRP-1)和P物质表达的积分光密度值(IOD)。结果偏治颗粒中、高剂量组及对照组大鼠耳红出现时间较模型组增加(P<0.05),耳红消失时间较模型组缩短(P<0.05)。模型组大鼠CGRP-1和P物质表达的IOD均高于空白组(P<0.05)。偏治颗粒中、高剂量组及对照组CGRP-1表达的IOD均低于模型组(P<0.05),偏治颗粒高剂量组及对照组P物质表达的IOD均低于模型组(P<0.05)。结论偏治颗粒可通过调节CGRP-1和P物质表达对硝酸甘油诱导的偏头痛大鼠模型产生保护作用。Objective To observe the Preventive effect and mechanism of Pianzhi granules on nitroglycerin-induced migraine in rat model.Methods 60 SD rats were divided into 6 groups according to the random number table method:blank group,model group,low dose group(5.4 g crude drug/kg),middle dose group(10.8 g crude drug/kg),high dose group(20.16 g crude drug/kg)and control group(18 mg/kg of sumaptan succinate tablets).Rats in each group were given prophylactic drug once a day for 7 days,with a volume of 10 mL/(kg·d).Distilled water of equal volume was given to blank group and model group.One hour after the last administration,except for the blank group,the other five groups were subcutaneously injected with nitroglycerin 10 mg/kg to duplicate the migraine model.The appearance and disappearance time of auricular red in each group were observed,and the expression of calcitonin gene related peptide type 1(CGRP-1)in spinal trigeminal tract nucleus and integrated optical density(IOD)of substance P in spinal trigeminal tract nucleus of each group were compared.Results Compared with the model group,the appearance time of auricular red in the middle and high dose group of Pianzhi granules and the control group was increased(P<0.05),and the disappearance time of auricular red was decreased(P<0.05).The IOD values of CGRP-1 and substance P in the model group were higher than those in the blank group(P<0.05).The IOD value of CGRP-1 expression in the middle and high dose group of Pianzhi granules and the control group was lower than that in the model group(P<0.05),and the IOD value of substance P expression in the high dose group of Pianzhi granules and the control group was lower than that in the model group(P<0.05).Conclusion Pianzhi granules can protect migraine rat model induced by nitroglycerin by regulating the expression of CGRP-1 and substance P.
