摘  要:目的探讨转化生长因子-α(TGF-α)、转化生长因子-β(TGF-β1)和微小RNA-302a(miR-302a)在胃癌患者组织中的表达及其与胃癌的相关性。方法选取2016年6月至2018年6月唐山市人民医院收治的92例胃癌患者作为研究对象,患者均接受手术治疗,手术过程中取病灶组织,设为胃癌组;选择癌旁组织(距离病灶≥3 cm),设为对照组。采用实时荧光PCR检测技术测定92组织样本中miR-302a的表达量,同时采用免疫组化法检测组织样本中TGF-α和TGF-β1的表达水平;并分析其与胃癌分级、淋巴转移和TNM分期等的相关性。结果胃癌组织中TGF-β1呈强阳性表达,在胃癌组织旁增殖区域及旺盛增殖区域呈现弱阳性表达,在正常黏膜中呈现弱阳性或阴性表达。胃癌组中TGF-α和TGF-β1表达水平明显高于对照组(P<0.01),胃癌组miR-302a表达水平明显低于对照组(P<0.01)。胃癌组织中TGF-α、TGF-β1和miR-302a表达水平与年龄、性别无统计意义(P>0.05);胃癌组织中TGF-α、TGF-β1和miR-302a表达水平与组织分化程度、淋巴结转移、TNM分期相关(P>0.05)。胃癌组织中TGF-α、TGF-β1和miR-302a表达水平与患者临床分期呈正相关性(P<0.05)。结论TGF-α、TGF-β1和miR-302a在胃癌组织中表达量显著改变,且与肿瘤细胞的TNM分期、分化程度和淋巴结转移相关,提示TGF-α、TGF-β1和miR-302a可能在胃癌的发生、分化和转移过程中起一定作用。Objective To investigate the expression of transforming growth factor-α(TGF-α),transforming growth factor-β(TGF-β1)and microRNA-302a(miR-302a)in tissues of gastric cancer patients and their relationship with gastric cancer Correlation.Methods A total of 92 patients with gastric cancer were enrolled in our hospital from June 2016 to June 2018.All patients underwent surgery.The lesions were taken during the operation and were set as gastric cancer.The adjacent tissues were selected(distance lesion≥3 cm),set as the control group.Real-time fluorescent PCR was used to detect the expression of miR-302a in 124 tissue samples.The expression levels of TGF-αand TGF-β1 in tissue samples were detected by immunohistochemistry.The lymphocyte metastasis and lymphatic metastasis were analyzed.Correlation with TNM staging,etc.Results TGF-β1 was strongly expressed in gastric cancer tissues,and weakly positively expressed in proliferative areas and proliferating areas adjacent to gastric cancer tissues,and weakly positive or negative in distant and normal mucosa.The expression levels of TGF-αand TGF-β1 in gastric cancer group were significantly higher than those in control group(P=0.000).The expression level of miR-302a in gastric cancer group was significantly lower than that in control group(P=0.000).TGF-αand TGF-in gastric cancer tissues.The expression levels ofβ1 and miR-302a were not statistically significant(P>0.05).The expression levels of TGF-α,TGF-β1 and miR-302a in gastric cancer tissues were significantly different from those of tissue differentiation,lymph node metastasis and TNM staging.(P>0.05);SPSS Pearson correlation analysis showed that the expression levels of TGF-α,TGF-β1 and miR-302a in gastric cancer tissues were positively correlated with clinical stage(P<0.05).Conclusion The expression levels of TGF-α,TGF-β1 and miR-302a in gastric cancer tissues are significantly changed,which are related to the TNM stage,differentiation degree and lymph node metastasis of tumor cells,suggesting that TGF-�
