摘 要:目的比较不同成份脱钙液对骨组织脱钙的效果。方法选取2019年3—10月在四川省八一康复中心手术室送检的股骨头标本20例为研究对象。将新鲜离体股骨头组织制成大小约1.0 cm×1.0 cm×0.3 cm标本,所有骨组织均分成3份,分别放入甲、乙、丙3种新鲜脱钙液中(甲液:10%硝酸甲醛液;乙液:30%盐酸甲醛液;丙液:30%盐酸甲酸甲醛液),在常温下进行脱钙,记录脱钙时间,观察3组标本切片质量、HE染色和免疫组化染色效果。结果3组脱钙时间比较差异有统计学意义(P<0.05)。与甲组相比,乙组和丙组脱钙时间明显缩短(P<0.05),并且丙组脱钙时间比乙组短(P<0.05)。丙组的骨组织经HE及免疫组化染色后,细胞核和细胞质着色均匀、清晰,对比明显、易于观察。结论30%盐酸甲酸甲醛液能缩短股骨头组织的脱钙时间,切片质量好,染色效果佳。Objective To compare the decalcification effect of different decalcification solutions on bone tissue.Methods A total of 20 femoral head specimens sent to be detected from the operating room of Sichuan Bayi Rehabilitation Center from March to October 2019 were selected as research objects.The freshly isolated femoral head tissues were made into specimens of about 1.0cm×1.0cm×0.3cm.All the bone tissues were divided into three parts and put into three kinds of newly prepared decalcification solutions(10%formaldehyde nitrate solution for group A,30%formaldehyde hydrochloride solution for group B,and 30%hydrochloric and formic acid formaldehyde solution for group C,respectively)at room temperature for decalcification.The decalcification time was recorded.The slice quality and the effect of HE staining and immunohistochemical staining were observed.Results There were significant differences in the decalcification time between the three groups(P<0.05).The decalcification time was significantly shorter in group B and group C than in group A(P<0.05).And the decalcification time of group C was shorter than that of group B(P<0.05).The bone tissues of group C were stained by HE and immunohistochemical staining.And the nuclei and cytoplasm were stained uniformly and clearly,with obvious contrast,which was easy to observe.Conclusion 30%hydrochloric and formic acid formaldehyde solution can shorten the decalcification time of femoral head tissue.The section quality is good,and the staining effect is good.
