摘　　要：目的:探讨白介素-1β(IL-1β)基因-511多态性位点与精神分裂症(SZ)及其临床特征的相关性。方法:采用TaqMan探针基因分型技术对320例SZ患者(SZ组)和284名健康对照者(对照组)的IL-1β基因-511位点进行基因分型,比较两组的等位基因、基因型频率分布差异。采用阳性和阴性症状量表(PANSS)评估患者的精神病性症状,比较-511位点不同基因型患者症状的严重程度。结果:两组IL-1β基因-511位点基因型分布均符合Hardy-Weinberg平衡(χ^2值=1.50,3.18,P均>0.05);两组-511位点GG、GA和AA基因型分布以及G、A等位基因频率分布比较差异无统计学意义(P>0.05);携带A等位基因的患者阴性症状分(F=43.72,P<0.001)和PANSS总分(F=10.53,P=0.001)显著高于不携带A等位基因的患者,两组阳性症状分和一般精神病理症状分比较差异无统计学意义(P>0.05)。结论:IL-1β基因-511位点可能与SZ发病无关,但与疾病的阴性症状严重程度有关。Objective:To investigate the relationship between interleukin-1β(IL-1β)gene-511 polymorphism and schizophrenia(SZ)and its clinical features.Method:TaqMan probe genotyping technique was used to genotype the IL-1βgene-511 locus in 320 SZ patients(SZ group)and 284 healthy controls(control group).The differences in genotype and alleles distribution frequency between the two groups were compared.Positive and Negative Syndrome Scale(PANSS)was used to assess psychiatric symptoms in the SZ patients,and the severity of disease symptoms between different genotypes of-511 locus were compared.Results:The IL-1βgene-511 genotype distribution was consistent with the Hardy-Weinberg equilibrium in both groups(χ^2=1.50,3.18,P>0.05).The differences in distribution of GG,GA and AA genotypes and the frequency distribution of G and A alleles were not statistically significant(P>0.05).Patients with A allele had higher negative gene score(F=43.72,P<0.001)and PANSS total score(F=10.53,P=0.001)than those without A allele,but there were no significant differences between the positive symptom scores and the general psychopathological symptoms between the two groups(P>0.05).Conclusion:The IL-1βgene-511 polymorphism may not be associated with schizophrenia,but it is associated with the severity of the negative symptoms of the disease.
