摘 要:目的:对比分析碘营养适宜和过量对有自身免疫性甲状腺炎遗传倾向小鼠甲状腺转录组的影响,为碘过量致自身免疫性甲状腺疾病的发病机制提供科学依据。方法:将18只5周龄NOD·H-2^h4小鼠随机分为对照组和高碘组,分别予适宜碘和过量碘饲养16周。ELISA和HE法进行血清甲功和甲状腺病理检测。建立甲状腺转录组文库,利用Illumina HiSeq平台测序,进行基因转录组表达差异和差异基因KEGG通路分析。结果:适宜碘和过量碘饲养建立了甲状腺正常和自身免疫性甲状腺炎小鼠模型,两组小鼠甲状腺转录组存在304个差异表达基因,其中与细胞因子、细胞趋化因子和细胞粘附分子通路,以及抗原处理呈递相关的基因转录水平在高碘组显著升高。结论:碘过量可能会过度激活多个与免疫反应和炎症发生密切相关的分子通路,共同促进自身免疫性甲状腺疾病的发生发展。【Objective】To provide scientific basis for the molecular mechanism of iodine-induced autoimmune thyroiditis,we analyzed the effects of excessive and appropriate iodine on the transcriptome of thyroid gene in mice with genetic predisposition to autoimmune thyroiditis.【Method】Totally 18 NOD·H-2^h4 mice 5 week old were randomly divided into control group and high iodine group.The control group was given distilled water,while the high iodine group was given distilled water containing 0.05%NaI for continuous induction for 16 weeks.The serum thyroid function and thyroid pathology were detected by ELISA and HE staining.Illumina HiSeq platform was used for sequencing,and then the gene expression differences between different samples,and the Kyoto encyclopedia of genes and genomes(KEGG)were analyzed.【Result】The rat models of normal thyroid and autoimmune thyroiditis were established by appropriate and excessive iodine feeding.Totally 304 genes expressed differentially in the thyroid transcriptome between the control group and the high iodine group.The cytokine pathway,cytokinokine pathway,cell adhesion molecular pathway and antigen processing and presenting related genes were significantly increased in thyroid tissues of the high iodine group.【Conclusion】Excessive iodine intake may promote the occurrence and development of autoimmune thyroiditis by activating multiple molecular pathways closely related to immune response and inflammation.
