摘　　要：目的探讨微小RNA-149-5p(miRNA-149-5p)在弥漫大B细胞淋巴瘤(DLBCL)中的表达情况及作用机制。方法选择150例DLBCL患者和70例淋巴结反应性增生患者,分别作为观察组和对照组。采用逆转录聚合酶链法检测两组患者中miRNA-149-5p的相对表达量,采用免疫组织化学染色法检测两组患者中核因子κB(NF-κB)/p65、B细胞淋巴瘤/白血病-2(Bcl-2)、胱天蛋白酶3(caspase 3)、Ki-67蛋白的表达情况。依据miRNA-149-5p表达水平,将DLBCL患者分为miRNA-149-5p高表达组和miRNA-149-5p低表达组,比较两组患者的3年生存率。结果观察组患者的miRNA-149-5p相对表达量明显低于对照组,差异有统计学意义(P﹤0.01)。miRNA-149-5p表达情况可能与DLBCL患者的Hans分型有关(P﹤0.01)。观察组患者的NF-κB/p65、Bcl-2、Ki-67蛋白阳性率均明显高于对照组,caspase 3蛋白阳性率明显低于对照组,差异均有统计学意义(P﹤0.01)。miRNA-149-5p高表达组患者的3年生存率为46.4%,明显高于miRNA-149-5p低表达组的17.5%,差异有统计学意义(P﹤0.01)。结论 DLBCL患者中miRNA-149-5p表达下调,可能通过凋亡途径影响肿瘤的发生,miRNA-149-5p低表达可能与DLBCL患者预后不良有关。Objective To explore the expression and mechanism of action of microRNA-149-5p(miRNA-149-5p)in diffuse large B-cell lymphoma(DLBCL).Method A total of 150 patients with DLBCL and 70 patients with reactive lymphoid hyperplasia(RLH)were included in the analysis as study group and control group,respectively.The relative expression level of miRNA-149-5p in both groups was detected by reverse transcription-polymerase chain reaction.The expression of nuclear factor-κB(NF-κB)/p65,B cell lymphoma/leukemia-2(Bcl-2),caspase 3 and Ki-67 was detected by immunohistochemistry.The 3-year survival rates were compared for patients with high expression or low expression of miRNA-149-5p.Result Compared with control group,the expression of miRNA-149-5p was lower in study group(P<0.01).The expression of miRNA-149-5p in DLBCL patients might be related to Hans typing(P<0.01).The positive rates of NF-κB/p65,Bcl-2 and Ki-67 were higher in study group,while the positive rate of caspase 3 were lower in study group versus control group(P<0.01).The 3-year survival rate of patients with high miRNA-149-5p expression was 46.4%,compared to a significantly lower rate of 17.5%in those with low miRNA-149-5p expression(P<0.01).Conclusion The expression of miRNA-149-5p is down regulated in patients with DLBCL,which may be involved with tumorigenesis through the apoptosis pathway,low expression of miRNA-149-5p is likely related to the poor prognosis of patients with DLBCL.
