摘  要:目的研究长链非编码RNA(lncRNA)LOC554202作为内源竞争RNA(ceRNA)调控微小RNA(miR)-98-5p在卵巢癌顺铂(DDP)化疗敏感性中的作用。方法利用双荧光素酶检测、定量PCR和细胞计数试剂盒研究LOC554202和miR-98-5p之间的靶向关系,卵巢癌A2780细胞株和DDP耐药的卵巢癌A2780(A2780/DDP)细胞株中LOC554202和miR-98-5p表达及相互作用情况,LOC554202对A2780细胞株和A2780/DDP细胞株增殖和耐药的影响。结果与A2780细胞株比较,A2780/DDP细胞株中LOC554202低表达,而miR-98-5p高表达(P<0.05)。对于A2780和A2780/DDP细胞株而言pcDNA3.1-LOC554202组LOC554202相对表达量均高于pcDNA3.1组,而miR-98-5p相对表达量均低于pcDNA3.1组(P<0.05);siRNA-LOC554202组LOC554202相对表达量均低于siRNA-NC组,而miR-98-5p相对表达量均高于siRNA-NC组(P<0.05);LOC554202过表达均可显著抑制细胞增殖和降低细胞IC50,而LOC554202敲降均可显著促进细胞增殖和提升细胞IC50(P<0.05)。结论LOC554202和miR-98-5p可能与卵巢癌DDP耐药相关。LOC554202可作为ceRNA调控miR-98-5p的表达,进而影响卵巢癌DDP化疗的耐药。Objective To investigate the role of long non-coding RNA(lncRNA)LOC554202 as competing endogenous RNA(ceRNA)regulating micro RNA(miR)-98-5p in the cisplatin(DDP)chemosensitivity of ovarian cancer.Methods Luciferase reporter,quantitative PCR and CCK-8 were used to detect the relationship between LOC554202 and miR-98-5p,the expression and interaction of miR-98-5p and LOC554202 in ovarian cancer A2780 cell lines and DDP resistant ovarian cancer A2780(A2780/DDP)cell lines,and the effect of LOC554202 on the proliferation and IC50 of the A2780 cell lines and the A2780/DDP cell lines.Results Compared with the A2780 cell lines,LOC554202 expression was lower and miR-98-5p expression was higher in the A2780/DDP cell lines(P<0.05).The LOC554202 relative expression quantity in the pcDNA3.1-LOC554202 group were higher than those in the pcDNA3.1 group,but the miR-98-5p relative expression quantity in the pcDNA3.1-LOC554202 group were lower than those in the pcDNA3.1 group(P<0.05);the LOC554202 relative expression quantity in the siRNA-LOC554202 group were lower than those in the siRNA-NC group,but the miR-98-5p relative expression quantity in the siRNA-LOC554202 group were higher than those in the siRNA-NC group(P<0.05);high-expression of LOC554202 could significantly decrease cell proliferation and IC50,but low-expression of LOC554202 could significantly increase cell proliferation and IC50(P<0.05).Conclusion LOC554202 and miR-98-5p might be related to DDP resistance in ovarian cancer,LOC554202 regulates the expression of miR-98-5p by ceRNA and DDP resistance in ovarian cancer.
