摘　　要：目的比较长链非编码核糖核酸(lncRNA)GAS8-AS1在甲状腺乳头状癌患者术前和术后的表达水平,探讨lncRNA GAS8-AS1作为生物标志物用于甲状腺乳头状癌临床诊断的可行性。方法入组81例甲状腺乳头状癌患者作为试验组,66例结节性甲状腺肿患者作为对照组;采集术前和术后1 a的血液样本并收集术中的肿瘤组织,运用荧光定量逆转录聚合酶链反应(qRT-PCR)检测血液、肿瘤组织中lncRNA GAS8-AS1表达水平,比较术前和术后1 a血液、肿瘤组织中lncRNA GAS8-AS1表达的差异。结果术前,试验组患者血液中lncRNA GAS8-AS1相对表达量较对照组明显减少(P<0.05)。术前,试验组患者肿瘤组织中lncRNA GAS8-AS1相对表达量与其血液中的相对表达量相近(P>0.05),且较对照组血液中的相对表达量明显降低(P<0.05)。术后1 a,试验组患者血液中lncRNA GAS8-AS1的相对表达量较术前明显升高(P<0.05),且与对照组血液中的相对表达量相近(P>0.05)。结论lncRNA GAS8-AS1可以作为生物标志物在甲状腺乳头状癌的临床诊断和术后监测过程中起到关键作用。Objective To compare the expression levels of long non-coding ribonucleic acid(lncRNA)GAS8-AS1 in the papillary thyroid carcinoma patients before and after surgery,and to discuss the feasibility of lncRNA GAS8-AS1 as the biomarker for the clinical diagnosis of papillary thyroid carcinoma.Methods The 81 papillary thyroid carcinoma patients were collected,meanwhile,blood samples from patients before and after surgery,operative tumor tissues from patients were collected.The 66 nodular goiter patients were collected as control.The expression abundance and difference of lncRNA GAS8-AS1 were determined by quantitative reverse transcript polymerase chain reaction compared with the control group.Results The plasma expression of lncRNA GAS8-AS1 of the patients before surgery in the test group was significantly lower compared with the control group(P<0.05),meanwhile,the expression of the tumor tissues was similar as the plasma(P>0.05),but was remarkably reduced compared with the control group(P<0.05).In addition,the plasma expression of lncRNA GAS8-AS1 of the patients in the 1 year after surgery was increased compared with before surgery(P<0.05),and was also similar as the control group(P>0.05).Conclusion lncRNA GAS8-AS1 as the biomarker can play a key role in the clinic diagnosis and postoperative monitor for papillary thyroid carcinoma.
