摘  要:目的探讨3D生物打印的肝癌模型用于顺铂药敏试验的可行性。方法通过3D生物打印技术分别构建SMMC-7721和Hep G2细胞3D水凝胶肝癌模型,并对不同浓度的顺铂进行药敏试验,采用LIVE/DEAD细胞荧光染色观察药物敏感性。并与2D培养药敏试验比较。结果在打印的3D肝癌模型环境中,SMMC-7721和Hep G2细胞分别形成葡萄串状细胞球体和块状肿瘤球体。而在2D培养中形成单层细胞。打印的3D肝癌模型中SMMC-7721细胞和Hep G2细胞对顺铂的敏感性均显著低于2D培养模型(P<0.05)。结论3D打印的肿瘤模型可更真实地模拟体内肿瘤生长的微环境,有助于提高药敏试验的可靠性,可在细胞实验与临床实验的联系中发挥作用。Objective To explore the feasibility of 3D bio-printed liver cancer model and its susceptibility to cisplatin,3D printed tumor model was employed.Methods 3D hydrogel liver cancer models of SMMC-7721 and HepG2 cells were constructed by 3D bio-printing technology,and drug sensitivity tests of the liver cancer models to cisplatin were carried out by LIVE/DEAD cells fluorescence staining.Then the obtained results of the 3D printed model were compared with that of 2D culture model.Results SMMC-7721 and HepG2 cells formed grape cell spheres and massive tumor spheres respectively in the printed 3D liver cancer model environment while monolayer cells were formed in 2D culture.The sensitivity of SMMC-7721 cells and HepG2 cells to cisplatin in 3D model was significantly lower than that in 2D model(P<0.05).Conclusion The 3D printed tumor model can more realistically simulate the microenvironment of tumor growth in vivo,which is helpful to improve the reliability of drug sensitivity test and can play a role in the connection between cell experiment and clinical experiment.
