摘  要:目的:分析针刺对帕金森病(PD)模型小鼠肠道菌群多样性的影响,从对肠道菌群调节作用角度探讨针刺治疗PD的可能性机制。方法:采用MPTP腹腔注射的方法建立慢性PD小鼠模型,针刺组选取百会、阳陵泉进行干预,采用Miseq平台对各组小鼠粪便V3~V4区肠道菌群进行双端测序,对结果进行OTU聚类、Alpha多样性分析、LEfSe差异分析、不同组差异菌群显著性分析等。结果:针刺对于PD模型小鼠运动功能具有显著改善作用,且可以保护其中脑黑质神经元。针刺可以显著调节肠道菌群丰度,并可改善模型小鼠肠道微生物的多样性,上调拟杆菌、普氏菌以及粪杆菌等抗炎、保护肠神经的菌群,下调艾克曼菌等破坏肠道屏障的菌群。结论:针刺可能通过调节PD模型小鼠肠道菌群而起到神经保护作用。Objective:To investigate the protective effect of acupuncture on Parkinson’s disease(PD)and its possible mechanism through analyzing the influence of acupuncture on regulating the microbial diversity of the PD mice.Methods:The mice model of PD was established by intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP),Baihui(DU20)and Yinlingquan(SP9)were selected as acupuncture points in acupuncture group.Miseq platform was used to perform the high-throughput sequencing based on 16S rRNA gene V3〜V4 region of the PD mice,OTU cluster,Alpha diversity and linear discriminant analysis of the effect size(LEfSe),and the differences of intestinal microbial community between different groups of PD mice were performed.Results:Acupuncture could significantly improve the motor function of PD mice,and protect the substantia nigra neurons of midbrain.Acupuncture had a positive effect on regulating the abundance of intestinal microbiome in PD mice,improving the diversity of intestinal microbiome,up-regulating bacteria which was anti-inflammatory and protective to intestinal neurons,such as Bacteroidetes,Prevotellaceae and Verrucomicrobia,down-regulating bacteria which could damage intestinal barrier,such as Akkermansia.Conclusion:Acupuncture may have neuroprotective effect by regulating intestinal microbiome in PD mice.
