摘　　要：目的评估Notch配体Dll-4在膀胱移行细胞癌中的表达及临床意义。方法 70例膀胱移行细胞癌进行免疫组化检测评估Dll-4在膀胱癌标本中的表达情况,同时采用RT-PCR和Western blot检测肿瘤组织中目标基因及蛋白表达情况,10例正常膀胱镜体检组织作为对照。在蛋白和基因水平分析Dll-4表达同膀胱癌,肿瘤大小、级别、复发及转移、预后的关系,并且利用生物信息分析验证研究结果。结果 Dll-4在膀胱癌组织中检测,基因及蛋白水平表达均较空白对照组显著增高,免疫组化结果显示Dll-4在高级别肿瘤中表达较低级别明显增高,但在RT-PCR及Western-blot检查中未见明显差异,Dll-4高表达同肿瘤复发、转移无显著相关性。结论 Dll-4参与膀胱移行细胞癌的发生发展,同时Dll-4作为潜在的膀胱癌靶向治疗点值得进一步深入研究。Objective To evaluate the expression and clinical significance of Notch ligand Dll-4 in bladder transitional cell carcinoma.Methods 70 cases of bladder transitional cell carcinoma were detected by immunohistochemistry to evaluate the expression of Dll-4 in bladder cancer specimens.The expression of target gene and protein in tumor tissue was detected by RT-PCR and Western blot,and 10 cases of normal cystoscopy examination tissue as control.RT-PCR and Western blot were used to detect the expression of target genes and proteins in tumor tissues.Organization as a control.At the protein and gene levels,the relationship between Dll-4 expression and bladder cancer,tumor size,grade,recurrence and metastasis,and prognosis was analyzed,and bioinformatic analysis was used to verify the research results.Results The expression of Dll-4 in bladder cancer tissues was significantly higher than that in the blank control group.Immunohistochemistry showed that the expression of dll-4 was significantly higher in high-grade tumors than that in low-grade tumors,while in RT-PCR and there was no obvious difference in western-blot examination,and the high expression of Dll-4 had no significant correlation with tumor recurrence and metastasis.Conclusion Dll-4 could be involved in the occurrence and development of bladder transitional cell carcinoma.and it is worth further research as a potential target therapy for bladder cancer.
