摘  要:目的基于TGF-β1/Smads信号通路探讨胆通颗粒对胆汁淤积性早期肝纤维化大鼠的抗肝纤维化作用。方法将80只Wistar大鼠随机分为空白组、模型组、消炎利胆片组、胆通颗粒组,每组20只。除空白组大鼠外,其余组大鼠均采用胆总管结扎方法进行胆汁淤积性早期肝纤维化模型建立。末次造模后第1天开始,模型组大鼠灌胃生理盐水,消炎利胆片组大鼠灌胃消炎利胆片(研磨成粉,175 mg/kg,温水融化),胆通颗粒组大鼠灌胃胆通颗粒(200 mg/kg,温水融化),均1 mL/100 g。分别于灌胃1,5,10 d后,每组随机处死6只大鼠,应用ELISA法检测各组大鼠血清TGF-β1水平,Western Blot法检测肝脏组织中Smad2、Smad3蛋白表达水平。结果灌胃1,5,10 d后,模型组、消炎利胆片组、胆通颗粒组大鼠血清TGF-β1水平及肝脏组织中Smad2、Smad3蛋白相对表达水平均明显高于空白组(P均<0.05);灌胃5,10 d后,消炎利胆片组、胆通颗粒组大鼠血清TGF-β1水平及肝脏组织中Smad2、Smad3蛋白相对表达水平均明显低于模型组(P均<0.05);胆通颗粒组灌胃10 d后血清TGF-β1水平及灌胃5,10 d后肝脏组织中Smad3蛋白相对表达水平均明显低于消炎利胆片组(P均<0.05)。结论胆通颗粒能够通过降低TGF-β1水平及抑制Smad2、Smad3蛋白表达,从而阻断TGF-β1/Smads信号通路来实现抗胆汁淤积性早期肝纤维化的作用。Objective It is to investigate the anti-hepatic fibrosis effect of Dantong granule on TGF-β1/Smads signaling pathway in rats with early cholestatic hepatic fibrosis.Methods 80 Wistar rats were randomly divided into blank group,model group,Xiaoyanlidan tablet group and Dantong granule group,20 rats in each group.Except for the rats in the blank group,the rats in the other groups were used to established establish the models of early cholestatic liver fibrosis with the common bile duct ligation method.Beginning on the first day after the last modeling,the rats in the model group were given normal saline by gavage,the rats in the Xiaoyanlidan tablet group were given Xiaoyanlidan tablets(milled into powder,175 mg/kg,melted with warm water)by gavage,and the rats in the Dantong granule group were given Dantong granules(200 mg/kg,melted in warm water)by gavage,all 1 mL/100g.After 1,5,and 10 days of gavage,6 rats were randomly killed in each group.The levels of serum TGF-β1 of each group were detected by ELISA,and the levels of expression of Smad2 and Smad3 protein in liver tissue were detected by Western Blot method.Results After 1,5,10 days of gavage,the levels of serum TGF-β1 and the relative expression of Smad2 and Smad3 protein in liver tissue of the model group,Xiaoyanlidan tablet group and Dantong granule group were significantly higher than those of the blank group(P<0.05);after 5 and 10 days of intragastric administration,the levels of serum TGF-β1 and the relative expression of Smad2 and Smad3 protein in the liver tissues of Xiaoyanlidan tablet group and Dantong granule group were significantly lower than those of the model group(all P<0.05).The level of serum TGF-β1 after 10 days of gastric gavage and the level of Smad3 protein expression in liver tissue after 5,10 days of gastric gavage in Dantong granule group were significantly lower than those of Xiaoyanlidan tablet group(all P<0.05).Conclusion Dantong granule can block the TGF-β1/Smads signal pathway by decreasing the level of TGF-β1 and inhibiting
