摘  要:采用固体培养基平板分离纯化及测定对指示菌抑菌圈直径等方法筛选高产土霉素优良菌株,并对其土霉素生产的种子培养基和发酵培养基进行优化。实验结果表明,经分离纯化获得优良菌株B,其抑菌圈直径为27.25 mm;菌株B在未优化的发酵条件下,发酵8 d,效价达8943 U/mL,比原菌效价(6430 U/mL)提高39%;采用优化了的最适种子培养基1号和最适发酵培养基2号发酵8 d,土霉素效价达到10247 U/mL;菌株B在以经淀粉酶预处理的发酵培养基2号发酵6 d,效价最高达11023 U/mL,较原菌株效价提高71.43%。The excellent strains with high yielding oxytetracycline were screened by solid medium plate for separating,purifying and determining the diameter of bacteriostatic zone of indicator bacteria,and then the seed medium and fermentation medium for oxytetracycline production were optimized.The experimental results showed that the excellent strain B was obtained by isolation and purification,its inhibition zone diameter was 27.25mm.When strain B was fermented for 8 days under unoptimized fermentation conditions,the titer of oxytetracycline reached 8943 U/mL,which was 39%higher than that of the original strain(6430 U/mL).When strain B was fermented for 8 days with optimized seed medium No.1 and fermentation medium No.2,its titer was 10247 U/mL.When strain B was fermented for 6 days in the fermentation medium No.2 pretreated with amylase,its titer was as high as 11023 U/mL,which was 71.43%higher than that of the original strain.
