摘  要:目的探讨环状RNA-circEPSTI1在子宫颈癌生长与侵袭中的作用及其机制。方法采用qRT-PCR技术检测子宫颈癌细胞系及组织、正常子宫颈上皮细胞及正常子宫颈组织中circEPSTI1的表达,分析circEPSTI1的表达与子宫颈癌临床病理特征的关系;转染si-circEPSTI1干扰SiHa与HeLa细胞中circEPSTI1的表达,采用qRT-PCR技术验证转染效率。采用CCK-8、Transwell侵袭和裸鼠成瘤实验检测circEPSTI1对子宫颈癌细胞生长与侵袭的影响。采用Western blot法检测circEPSTI1对EPSTI1、Twist、Slug蛋白表达的影响。结果circEPSTI1在子宫颈癌细胞系中的表达水平明显高于正常子宫颈上皮细胞(P<0.01);circEPSTI1在子宫颈癌组织中的表达水平明显高于正常子宫颈组织(P<0.01);circEPSTI1表达与子宫颈癌的高级别FIGO分期、组织学分级以及淋巴结转移密切相关;qRT-PCR结果显示,SiHa与HeLa细胞中circEPSTI1被成功干扰。CCK-8实验显示,si-circEPSTI1组的SiHa与HeLa细胞的OD值均明显低于si-NC组(P<0.01)。在裸鼠成瘤实验中,si-circEPSTI1组的肿瘤质量明显低于si-NC组(P<0.05)。Transwell实验结果显示,si-circEPSTI1组的SiHa与HeLa细胞穿过基质胶的细胞数量均明显低于si-NC组(P<0.01)。Western blot法结果显示,si-circEPSTI1组的EPSTI1、Twist、Slug蛋白表达水平明显低于si-NC组(P<0.05)。结论circEPSTI1在子宫颈癌中表达增高并与子宫颈癌的高级别FIGO分期、组织学分级以及淋巴结转移密切相关,抑制circEPSTI1能够抑制子宫颈癌细胞的生长和侵袭,其机制可能与调控EPSTI1、Twist、Slug信号通路有关。Purpose This study is aimed to explore the role and mechanism of circular RNA-circEPSTI1 in the growth and invasion of cervical cancer.Methods qRT-PCR was used to detect the expression of circEPSTI1 in human cervical cancer cells and tissues,as well as normal cervical epithelial cells and normal cervical tissue.The relationship between the expression of circEPSTI1 in cervical cancer tissues and the clinicopathological characteristics was statistically analyzed.Transfection of si-circEPSTI1 knocked down the expression of circEPSTI1 in SiHa and HeLa cells,and the efficiency of transfection was verified by qRT-PCR.The effect of circEPSTI1 on the growth and invasion of cervical cancer cells was detected by CCK-8,Transwell invasion and mouse xenograft assay.Western blot was used to detect the effect of circEPSTI1 on the expression of EPSTI1,Twist and Slug proteins.Results The expression of circEPSTI1 in cervical cancer cells was significantly higher than that in normal cervical epithelial cells(P<0.01).The expression level of circEPSTI1 in cervical cancer tissues was higher than that in normal cervical tissues(P<0.01).Correlation analysis showed that the expression of circEPSTI1 was closely related to high-grade FIGO stage,histological grade and lymph node metastasis of cervical cancer patients.qRT-PCR showed that the si-circEPSTI1 was transfected successfully.CCK-8 results showed that the OD values of SiHa and HeLa cells in si-circEPSTI1 group were significantly lower than those in si-NC group(P<0.01).Mouse xenograft assay results showed that the tumor weight of si-circEPSTI1 group was significantly lower than that of si-NC group(P<0.05).Transwell results showed that the number of SiHa and HeLa cells passing through Matrigel in si-circEPSTI1 group were significantly lower than those in si-NC group(P<0.01).Western blot showed that the level of EPSTI1,Twist,Slug protein in si-circEPSTI1 group was significantly lower thanthat in si-NC group(P<0.01).Conclusion circEPSTI1 is highly expressed in cervical cancer cell lines
