摘　　要：目的探讨长链非编码RNA HOX转录反义RNA(HOTAIR)在卵巢上皮癌组织中的表达及与上皮钙黏素(E-cadherin)表达的相关性及临床意义。方法收集2015年1月至2018年12月在哈尔滨医科大学附属第一医院经手术治疗的45例卵巢上皮癌患者术中的新鲜组织标本,术后经病理证实浆液性卵巢上皮癌30例、黏液性卵巢上皮癌15例;另收集45例癌旁组织和手术治疗后经病理证实的45例良性卵巢上皮肿瘤组织及正常卵巢组织45例作为对照组。采用相对定量反转录聚合酶链反应法检测各组织中HOTAIR信使RNA(mRNA)的表达及免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连结法检测各组织中E-cadherin蛋白的表达,计算各组织中HOTAIR mRNA与E-cadherin蛋白的阳性表达率,分析HOTAIR与卵巢上皮癌临床病理特征的关系,并采用Spearman等级相关法分析卵巢上皮癌组织中HOTAIR mRNA与E-cadherin蛋白表达的相关性。结果 HOTAIR mRNA在卵巢上皮癌组织中相对表达量为4. 3±1. 7,在癌旁组织中相对表达量为0. 9±2. 2,在良性卵巢上皮肿瘤组织及正常卵巢组织中相对表达量均为0,不同组织间比较差异有统计学意义(P <0. 001)。HOTAIR mRNA在卵巢上皮癌组织、癌旁组织、良性卵巢上皮肿瘤组织、正常卵巢组织中的阳性率分别为82. 22%(37/45)、15. 56%(7/45)、0. 00%、0. 00%;E-cadherin蛋白在卵巢上皮癌组织、癌旁组织、良性卵巢上皮肿瘤组织、正常卵巢组织中的阳性率分别为84. 44%(38/45)、20. 00%(9/45)、2. 22%(1/45)、0. 00%,阳性表达主要位于细胞质。HOTAIR mRNA及E-cadherin蛋白在不同组织中的阳性率比较差异有统计学意义(P <0. 001)。卵巢上皮癌组织HOTAIR mRNA及E-cadherin蛋白的阳性率高于癌旁组织、良性卵巢肿瘤组织和正常卵巢组织,癌旁组织的阳性率高于良性卵巢肿瘤组织和正常卵巢组织(P <0. 05)。低分化组织的HOTAIR mRNA阳性率和相对表达量高于中分化和�Objective To investigate the correlation of the expression of long-chain noncoding RNA HOX transcriptional antisense RNA(HOTAIR)and the expression of E-cadherin in ovarian epithelial carcinoma tissues and its clinical significance.Methods Fresh tissue specimens were collected from 45 ovarian cancer patients who underwent surgical treatment in the First Affiliated Hospital of Harbin Medical University from Jan.2015 to Dec.2018,including 30 cases of serous ovarian epithelial carcinoma and 15 cases of mucous ovarian epithelial carcinoma confirmed by pathology after surgery.Additionally,45 paracancer ovarian tissues and 45 benign ovarian epithelial tumor tissues confirmed by pathology after surgical treatment and 45 normal ovarian tissues were collected as control groups.Relative quantitative reverse transcription polymerase chain reaction was used to detect the expression of HOTAIR messenger RNA(mRNA)in each tissue.Immunohistochemistry Streptavidin-peroxidase conjugate method was used to detect the expression of E-cadherin protein in each tissue.The HOTAIR mRNA and E-cadherin expression positive rates in each tissue was calculated,to analyze relationship between HOTAIR mRNA and the clinicopathological features of ovarian epithelial carcinoma,and the correlation between HOTAIR mRNA and E-cadherin expression in ovarian epithelial carcinoma tissues was analyzed by Spearman rank correlation method.Results The relative expression of HOTAIR mRNA in ovarian epithelial carcinoma tissues was(4.3±1.7),in paracancer tissues was(0.9±2.2),and in both benign ovarian epithelial tumor tissues and normal ovarian tissues was 0.0,with statistically significant differences between different tissues(P<0.001).The positive rates of HOTAIR mRNA in ovarian epithelial carcinoma,paracancer tissues,benign ovarian epithelial tumor and normal ovarian tissue were 82.22%(37/45),15.56%(7/45),0.00%and 0.00%,respectively.The positive rates of E-cadherin in ovarian epithelial carcinoma,paracancer tissues,benign ovarian epithelial neoplasm,and normal
