摘 要:为构建糯玉米品种DNA指纹库,保护品种资源,利用Illumina公司研发的包含56110个SNP标记的芯片,对50份糯玉米自交系进行基因分型。结果表明:按照SNP标记选用标准,共得到42406个SNP标记。这些标记的杂合率均值为0.04,SNP缺失率均值为0.01,最小等位基因频率(MAF)均值为0.4,多态性信息含量(PIC)均值为0.38,基因多样性指数均值为0.44。对上述获得的SNP标记设置不同数量位点进行筛选,确定25000位点为核心标记位点,可用于构建糯玉米自交系DNA指纹库。在25000位点时,对50份糯玉米自交系进行SNP聚类分析,共将其划分为四大类群,聚类结果与品种系谱来源一致。该研究可为糯玉米品种特异性和一致性的鉴定分析及品种保护工作提供理论指导。In order to construct the DNA fingerprint database of waxy corn varieties and protect the variety resources,50 waxy corn inbred lines were genotyped using a chip containing 56110 SNP(single nucleotide polymorphism)markers developed by Illumina.According to the selection criteria of SNP marker,a total of42406 SNP markers were obtained.The average heterozygosity rate was 0.04,the average SNP loss rate was 0.01,the average Minor Allelle Frequency(MAF)was 0.4,the average Polymorphic Information Content(PIC)was 0.38,the average gene diversity index was 0.44.Further,the number of core loci was screened by gradients analysis,and 25000 loci were optimal core loci for constructing fingerprinting of waxy corn inbred lines.Four groups were divided from the 50 inbred lines by cluster analysis,which was consistent with the pedigree.This study could provide guidances for the specificity,consistency of waxy corn varieties and varieties protection.
