摘  要:本试验旨在研究饲粮添加L-谷氨酰胺(L-Gln)对脂多糖(LPS)刺激肉鸡血浆生化指标、免疫性能、肠道炎症因子表达及黏膜免疫的影响。选择健康状况良好、体重相近的1日龄爱拔益加(AA)肉鸡120只,按2×2试验设计,随机分为4个组(A、B、C及D组),每组5个重复,每个重复6只鸡。A、C组饲喂对照饲粮(基础饲粮中添加1.22%L-丙氨酸),B、D组饲喂L-Gln饲粮(基础饲粮中添加1%L-Gln)。第16天及第21天,B、 D组肉鸡分别腹腔注射LPS(500μg/kg BW),A、C组肉鸡腹腔注射等量的生理盐水,注射前12 h停料不停水。试验期21 d。结果表明:1)LPS刺激显著增加了肉鸡血浆中总蛋白(TP)、尿酸(UA)含量及谷丙转氨酶(ALT)、谷草转氨酶(AST)活性(P<0.05),显著降低了胸腺指数、脾脏指数及法氏囊指数(P<0.05),显著降低了十二指肠相对长度、十二指肠相对重量,空肠相对长度、空肠相对重量、回肠相对长度及回肠相对重量(P<0.05),显著增加了十二指肠、空肠及回肠肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)及白细胞介素-6(IL-6)mRNA表达水平(P<0.05),显著增加了空肠黏膜分泌型免疫球蛋白A含量(P<0.05),显著增加了空肠黏膜黏蛋白1(MUC1)及黏蛋白2(MUC2)mRNA表达水平(P<0.05)。2)饲粮添加L-Gln显著降低了肉鸡TP、UA含量及ALT、AST活性(P<0.05),显著增加了胸腺指数、脾脏指数及法氏囊指数(P<0.05),显著增加了十二指肠相对长度、十二指肠相对重量,空肠相对长度、空肠相对重量、回肠相对长度及回肠相对重量(P<0.05),显著降低了十二指肠TNF-α及IL-6 mRNA表达水平(P<0.05),显著降低了空肠及回肠TNF-α、IL-1βmRNA表达水平(P <0. 05),显著增加了空肠黏膜sIgA含量(P<0.05),显著降低了空肠黏膜MUC1及MUC2 mRNA表达水平(P<0.05)。综上所述,饲粮中添加1%L-Gln能够提高肉鸡免疫性能,增加肠道黏膜免疫机能,从而在一定程度上有效缓解LPS刺激引起的肉鸡肠道炎症反�The present study aimed to investigate the effects of dietary L⁃glutamine(L⁃Gln)supplementation on plasma biochemical parameters,immune performance,intestinal inflammatory factors expression and mucosal immune of broilers by lipopolysaccharide(LPS)challenge.A total of 120 one⁃day⁃old Arbor Acres(AA)broilers with good health and similar initial body weight were arranged in a 2×2 factorial design to 4 groups(groups A,B,C and D)with 5 replicates per group and 6 broilers per replicate.Broilers in groups A and C were fed the control diets(basal diet supplemented with 1.22%alanine),and broilers in groups B and D were fed the L⁃Gln diets(basal diet supplemented with 1%L⁃Gln).At day 16 and day 21,broilers in groups B and D were administered with LPS(500μg/kg BW)by intraperitoneal injection,and broilers in groups B and D were administered with equivalent saline by intraperitoneal injection.The diet rather than water was cut off be⁃fore 12 h of injection.The experiment lasted for 21 days.The results were as follows:1)LPS challenge signif⁃icantly increased the total protein(TP),uric acid(UA)contents and alanine aminotransferase(ALT),aspar⁃tate transaminase(AST)activities in plasma of broilers(P<0.05),significantly decreased the thymus index,spleen index and bursa of Fabricius index(P<0.05),significantly decreased the relative length of duodenum,relative weight of duodenum,relative length of jejunum,relative weight of jejunum,relative length of ileum and relative weight of ileum(P<0.05),significantly increased the mRNA expression levels of tumor necrosis factor⁃α(TNF⁃α),interleukin⁃1β(IL⁃1β)and interleukin⁃6(IL⁃6)in duodenum,jejunum and ileum(P<0.05),significantly increased the content of secretory immunoglobulin A(sIgA)in jejunal mucosa(P<0.05),and significantly increased the mRNA expression levels of mucin 1(MUC1)and mucin 2(MUC2)in jejunal mucosa(P<0.05).2)Dietary L⁃Gln supplementation significantly decreased the TP,UA contents and ALT,AST activities in plasma of broilers(P<0.05),sign
