摘 要:本文建立了一种高效液相色谱(High performance liquid chromatography,HPLC)-紫外法测定畜禽肉中氟苯尼考残留量的方法。样品经乙腈提取,浓缩至干,加2 mL水复溶,用正己烷除脂净化,过0.22μm滤膜,高效液相色谱仪分析。色谱条件:色谱柱为C18柱,流动相为甲醇-水(40∶60),柱温为40℃,流速为0.2 mL·min^-1,紫外检测器在230 nm检测。本方法在10~500 ng·mL^-1范围内具有良好的线性关系,相关系数为0.999 5,在4个加标水平下,平均加标回收率为91.8%~95.2%,相对标准偏差为0.44%~3.58%。检出限为10μg·kg^-1,定量限30μg·kg^-1。该方法操作简单,结果准确,适合畜禽肉中氟苯尼考残留的检测。An analytical method based on HPLC-UV has been developed for determination of florfenicol in livestock and poultry muscle.The sample extract was concentrated up to dryness after extracted with acetonitrile and then dissolved in water with N-hexane purified,the aqueous solution was taken and analyzed by 0.22μm filter membrane and HPLC.Chromatographic conditions:chromatographic column was C18,mobile phase was methanol-water(40∶60),column temperature was 40℃,flow rate was 0.2 mL·min^-1,detected by UV-detector at 230 nm.The concentration and response of each component showed a good linear relationship in the range of 0~500 ng·mL^-1 with correlation higher than 0.9995.The average recoveries and relative standard deviations(RSD)are in the range of 91.8%~95.2%and 0.44%~3.58%at four spiked levels.The limits of detection and the limits of quantitative are in the range of 10μg·kg^-1 and 30μg·kg-1.The method is simple,accurate,and which is suitable for determination of florfenicol in livestock and poultry muscle.
