摘  要:目的分析MRN复合体的MRE11、RAD50和NBS13种蛋白表达与口腔鳞状细胞癌(OSCC)临床病理特征的关系。方法选取2016年1月-2017年6月广西医科大学附属口腔医院行OSCC根治术切取的癌组织标本作为观察组(50例),癌旁组织及良性疾病组织作为对照组(50例)。采用免疫组织化学SP法检测MRE11、RAD50和NBS1的表达。结果观察组MRE11、RAD50和NBS1阳性表达率分别为34%、38%和60%,对照组分别为56%、60%和32%,两组比较,差异有统计学意义(P<0.05),观察组MRE11、RAD50阳性表达率低于对照组,观察组NBS1的阳性表达率高于对照组(P<0.05);观察组NBS1的表达率在OSCC不同组织学分型、淋巴结有无转移和临床分期中比较,差异有统计学意义(P<0.05),NBS1在低、中分化组的阳性表达率高于高分化组(P<0.05),NBS1在有淋巴结转移患者的阳性表达率高于无淋巴结转移者(P<0.05),NBS1在Ⅲ、Ⅳ期患者的阳性表达率高于Ⅰ、Ⅱ期患者;观察组MRE11和RAD50的阳性表达率在OSCC不同分化程度和临床分期等临床特征中比较,差异无统计学意义(P>0.05)。结论MRE11、RAD50和NBS1阳性表达率在OSCC组织中存在异常,NBS1与OSCC的分化程度、淋巴结转移和临床分期有关。Objective To detect the expression of MRN complex in oral squamous cell carcinoma and analyze its correlation with the pathological features.Methods Fifty patients with oral cancer radical surgery at the Affiliated Stomatological Hospital of Guangxi Medical University(from January 2016 to June 2017)were enrolled as observation group,and the control group consisted of 50 cases of paracancerous or benign diseases.The expression of MRE11,RAD50 and NBS1 was detected by immunohistochemistry.Results The positive expression rates of MRE11,RAD50 and NBS1 in the observation group were 34%,38%and 60%,respectively.The control group was 56%,60%and 32%respectively.The difference between the two groups was statistically significantly(P<0.05).The positive rates of Mre11 and Rad50 in the observation group were lower than those in the control group,and the positive expression rate of NBS1 in the observation group was significantly higher than that in the control group(P<0.05).The positive expression rate of NBS1 in different OSCC tissue,lymphnode metastasis or not and clinical TNM stages was significantly different(P<0.05).The positive expression rate of NBS1 in low differentiation group and middle differentiation group was higher than that in high differentiation group(P<0.05);the positive expression rate of NBS1 in patients with lymph node metastasis was higher than that in patients without metastasis(P<0.05);the positive expression rate of NBS1 in clinical stageⅢandⅣwas higher than that in stageⅠandⅡ(P<0.05).There was no significant difference between Mre11 and Rad50 in different OSCC and TNM stages(P>0.05).Conclusions The positive expression rates of MRE11,RAD50 and NBS1 were different in OSCC and control tissues.NBS1 is related to the degree of OSCC differentiation,lymphnode metastasis and clinical TNM stage.
