摘  要:茎柔鱼广泛分布在东太平洋海域,分布水域广,种群结构复杂。采用线粒体COⅠ序列为遗传标记分析了东南太平洋茎柔鱼(Dosidicus gigas)遗传结构特征。在秘鲁外海8个群体239个样本的线粒体COⅠ序列中共检测到46种单倍型,42个变异位点。8个群体的单倍型多样性为(0.469±0.114)^(0.759±0.086),核苷酸多样性为(0.00104±0.00107)^(0.00363±0.00121),均具有较高的单倍型多样性水平和较低的核苷酸多样性水平。基于单倍型构建的NJ树以及基于群体间遗传距离构建的UPGMA树分析显示,8个群体间没有明显的地理谱系结构特征。AMOVA及F st分析结果表明,群体间变异百分比为0.26%,群体内变异百分比为99.74%,说明遗传变异主要来自群体内部,群体间不存在显著的遗传结构分化。群体间的基因流数据分析表明,各群体间具有显著的基因交流。研究结果可为茎柔鱼资源的合理利用、开发及科学管理提供必要参考。The jumbo squid Dosidicus gigas is an oceanic shallow water species and the largest cephalopod individual.It is a commercially important fishery species in the Eastern Pacific and is the most abundant in Peruvian waters.In order to exploit and manage this squid scientifically,we made a study of genetic population structure of D.gigas.Mitochondrial COI sequences were amplified by PCR and then used for genetic structure analysis.In the 239 individuals of 8 sampling locations,46 haplotypes were detected,and the haplotype diversity index and the nucleotide diversity index ranged from(0.469±0.114)to(0.759±0.086)and from(0.00104±0.00107)to(0.00363±0.00121),respectively.The results indicated that the haplotype diversity index(H d)of D.gagas in the high seas of the Southeastern Pacific was pretty high,while the nucleotide diversity index(π)was low.The shallow topology of the haplotype neighbor-joining(NJ)tree and the UPGMA tree based on genetic distance suggested that there was no distinct phylogeographic structure across the 8 populations.The results of AMOVA revealed that 0.26%of the genetic variation occurred between populations,and 99.74%occurred within populations.Fixation index(F st)analysis suggested that there was no significant genetic differentiation between the 8 populations.The estimate of gene flow between the populations was>1,indicating that gene exchanges among populations were frequent.Based on the analysis results,it can be speculated that there is no significant genetic differentiation because of the lack of geographical barriers in Peruvian Ocean,the role of Peru current and rip current,seasonal reproductive migration and strong swimming ability of D.gigas.
