摘 要:目的分析HMGA2基因对急性髓细胞白血病(AML)耐药细胞株HL-60/DNR对柔红霉素(DNR)耐药性的影响及机制。方法荧光定量PCR和western blot检测HMGA2的表达。通过转染HMGA2 siRNA(si-HMGA2)沉默HMGA2。CCK8实验检测DNR对HL-60/DNR细胞的抑制率。β-半乳糖苷酶活性检测试剂盒检测β-半乳糖苷酶活性。结果与野生型HL-60相比,HL-60/DNR中HMGA2 mRNA和蛋白的表达水平明显提高。DNR对cell组和si-NC组HL-60/DNR细胞的抑制率无明显差异。与si-NC组相比,DNR对si-HMGA2组HL-60/DNR细胞的抑制率显著提高。DNR对cell组、si-NC组和si-HMGA2组HL-60/DNR细胞的IC50分别为4.46μg/mL、4.67μg/mL和0.98μg/mL。cell组和si-NC组的β-半乳糖苷酶活性无明显差异;与si-NC组相比,si-HMGA2组HL-60/DNR细胞的β-半乳糖苷酶活性显著提高。结论HMGA2在耐药细胞HL-60/DNR中高表达,沉默HMGA2可以逆转HL-60/DNR的DNR耐药性并加速细胞的衰老。Objective To analyze the effect and mechanism of HMGA2 gene on drug resistance of acute myeloid leukemia(AML)cell line HL-60/DNR to daunorubicin(DNR).Methods Fluorescence quantitative PCR and western blot were used to detect the expression of HMGA2.Silencing HMGA2 by transfecting HMGA2 siRNA(si-HMGA2).CCK8 assay was used to detect the inhibition rate of DNR on HL-60/DNR cells.Theβ-galactosidase activity detection kit was used to detectβ-galactosidase activity.Results Compared to wild-type HL-60,the expression levels of HMGA2 mRNA and protein in HL-60/DNR were significantly increased.There was no significant difference in the inhibition rate of DNR on HL-60/DNR cells in cell group and si-NC group.The inhibition rate of DNR on HL-60/DNR cells was significantly increased in the si-HMGA2 group compared to that the si-NC group.The IC50 of DNR for HL-60/DNR cells in cell group,si-NC group and si-HMGA2 group were 4.46μg/mL,4.67μg/mL and 0.98μg/mL,respectively.There was no significant difference inβ-galactosidase activity between cell group and si-NC group;group.β-galactosidase activity of HL-60/DNR cells in si-HMGA2 group was significantly increased compared to that insi-NC group.Conclusion HMGA2 is highly expressed in drug-resistant cells HL-60/DNR.Silencing HMGA2 can reverse the DNR resistance of HL-60/DNR and accelerate cell senescence.
