摘  要:目的探讨白杨素(chrysin,CHR)对围绝经期抑郁症(perimenopausal depression disorder,PDD)大鼠的保护作用。方法将成功摘除双侧卵巢的大鼠随机分为空白对照组、模型组、CHR组(20、40、80 mg·kg^-1)。除假手术组外,其余各组大鼠进行每天随机接受慢性不可预见性应激刺激方式造模,持续5周。造模结束后,各组大鼠分别进行旷场实验、强迫游泳实验和糖水偏好实验,观察大鼠的行为学变化。采用HE染色观察大鼠海马神经元的变化,并采用Western blot法检测大鼠海马中caspase-1、NLRP3、ASC以及炎症因子的表达情况。结果与对照组相比,模型组大鼠单位时间内穿越格子数和站立次数均有所减少、糖水偏嗜度降低、强迫游泳不动时间延长;此外,海马神经元产生明显损伤,海马中caspase-1、NLRP3、ASC蛋白表达水平明显升高,炎症因子IL-1β、IL-18、IL-6达水平明显升高。与模型组相比,CHR药物治疗后大鼠单位时间内穿越格子数和站立次数均有所增加、糖水偏嗜度升高、强迫游泳不动时间缩短;此外,海马神经元产生损伤减少,海马中caspase-1、NLRP3、ASC蛋白表达水平降低,炎症因子IL-1β、IL-18、IL-6的表达水平明显降低。结论 CHR对PDD模型大鼠具有保护作用。Aim To explore the protective effect of chrysin(CHR)on perimenopausal depression disorder(PDD)in rats and its mechanisms.Methods Rats with bilateral ovariectomy(OVX)successfully removed were randomly divided into blank control group,model group,and CHR group(20,40,80 mg·kg^-1).Except for the sham operation group,the rats in the other groups were randomly and daily subjected to chronic unpredicted mild stress(CUMS)stimulation for 5 weeks.After the modeling was completed,rats in each group were subjected to open field experiments,forced swimming experiments,and sugar water preference experiments to observe the behavioral changes of the rats.The changes of rat hippocampal neurons were observed with HE staining,and the expressions of caspase-1,NLRP3,ASC and inflammatory factors in rat hippocampus were detected by Western blot.Results Compared with control group,the number of traversing lattices and standing times of the rats in model group were reduced,the sugar and water partiality was reduced,and the duration of forced swimming was significantly longer.The expression levels of caspase-1,NLRP3 and ASC proteins significantly increased,and the levels of inflammatory factors IL-1β,IL-18 and IL-6 significantly increased.Compared with model group,the number of crossing lattices and standing times per unit time increased in rats after CHR treatment,sugar and water partiality increased,and the time of forced swimming was shortened.In addition,damage to hippocampal neurons was reduced.The expression levels of caspase-1,NLRP3 and ASC proteins were down-regulated,and the expression levels of inflammatory factors IL-1β,IL-18 and IL-6 were significantly reduced.Conclusion Chrysin has neuroprotective effects on model rats with perimenopausal depression disorder.
