摘 要:目的研究结肠癌转移相关基因1(metastasis-associated in colon cancer 1,MACC1)对结肠癌细胞HT-29所致裸鼠结肠癌术后复发及肝转移的影响。方法采用shRNA法敲降MACC1表达。免疫荧光标记MACC1表达。噻唑蓝(thiazolyl blue,MTT)法检测细胞增殖能力。移植HT-29细胞建立裸鼠结肠癌复发和肝转移模型。结果shRNA敲降MACC1表达后,HT-29细胞中MACC1 mRNA和蛋白表达均下降,细胞增殖率降低45.2%(均P<0.05)。与移植HT-29细胞的对照组裸鼠比较,移植shRNA的细胞组裸鼠的移植瘤切除8周后复发肿瘤直径及重量分别降低55.8%和75.3%(均P<0.01)。与移植HT-29细胞对照组裸鼠比较,移植shRNA的细胞组裸鼠7周后肝转移形成的肿瘤结节数量、直径和融合面积均降低(均P<0.01)。结论敲降结肠癌细胞HT-29的MACC1可抑制HT-29结肠癌细胞的增殖,减少结肠癌模型裸鼠的复发和肝转移。Objective To study the effect of metastasis-associated in colon cancer l(MACC1)on the recurrence and liver metastasis of colon cancer in nude mice induced by HT-29 cells.Methods The expression of MACC1 was knocked down by sh RNA.The MACC1 expression was labeled by immunofluorescence.Cell proliferation was detected by thiazolyl blue assay.Recurrence and liver metastasis of colon cancer nude mice model were established by HT-29 cell transplantation.Results Compared with HT-29 cells,the mRNA and protein expression of MACC1 decreased significantly and the proliferation rate decreased 45.2% in the HT-29 cells transfected with shRNA(all P<0.05).After 8 weeks of tumor resection,compared with nude mice transplanted with HT-29 cells(control group),the diameter and weight of recurrent tumors decreased by 55.8% and 75.3%(both P<0.01),respectively,in the nude mice transplanted with shRNA transfected HT-29 cells(HT-29+shRNA-2 group).After 7 weeks of transplantation,compared with the control group,the number,size and fusion area of hepatic metastases were significantly reduced in the HT-29+shRNA-2 group(all P<0.01).Conclusion Knocking down MACC1 can inhibit the proliferation of colon cancer HT-29 cells and reduce the recurrence and liver metastasis of colon cancer model in nude mice.
