摘 要:目的测定毛菊苣不同部位(根、茎、种子)中菊苣酸、绿原酸和单咖啡酰酒石酸的含量。方法采用依利特Hypersil ODS柱(250 mm×4.6 mm,5μm),以乙腈-0.3%磷酸为流动相,梯度洗脱,流速为1.0 mL/min,检测波长为328 nm,柱温为35℃,进样量为10μL。结果菊苣酸、绿原酸、单咖啡酰酒石酸分别在3.22~51.60、1.33~42.56、0.256~8.208μg/mL浓度范围内线性关系良好;平均回收率分别为93.0%、101.3%、105.6%,RSD分别为3.0%、3.6%、2.6%。毛菊苣根中单咖啡酰酒石酸、绿原酸、菊苣酸的含量分别为0.024、0.092、0.143 mg/g;茎中单咖啡酰酒石酸、绿原酸、菊苣酸的含量分别为0.032、0.058、0.287 mg/g;种子中单咖啡酰酒石酸、绿原酸、菊苣酸的含量分别为0.197、1.714、1.247 mg/g。结论本法简单方便、重复性好、准确可靠,可用于毛菊苣的质量控制。Objective To determine the contents of cichoric acid,chlorogenic acid and monocaffeic tartaric acid in different parts(roots,stems and seeds)of Cichorium glandulosum Boiss.et Huet.Methods An Elite Hypersil ODS column(250 mm×4.6 mm,5μm)was used with acetonitrile-0.3%phosphoric acid as mobile phase.The gradient was eluted at a flow rate of 1.0 mL/min.The detection wavelength was 328 nm and the column temperature was 35℃.The injection volume was 10μL.Results Chicory acid,chlorogenic acid and caffeinyl tartaric acid have good linear relationship in the concentration range of 3.22-51.60,1.33-42.56 and 0.256-8.208μg/mL respectively.The average recovery is 93.0%,101.3%,105.6%and RSD is 3.0%,3.6%and 2.6%respectively.The contents of caffeic acid,chlorogenic acid and cichoric acid in the roots of chicory were 0.024,0.092 and 0.143 mg/g respectively;the contents of caffeic acid,chlorogenic acid and cichoric acid in the stems were 0.032,0.058 and 0.287 mg/g respectively;the contents of caffeic acid,chlorogenic acid and cichoric acid in the seeds were 0.197,1.714 and 1.247 mg/g respectively.Conclusion This method is simple,convenient,reproducible,accurate and reliable,and can be used for quality control of chicory.
