摘 要:目的探讨长链非编码RNA SNHG17(Lnc RNA SNHG17)和微小RNA-375(miR-375)在结直肠癌中的表达及意义。方法选取68例结直肠癌患者为结直肠癌组,留取术前及术后3个月第1次入院复查时的血样;70例健康体检者为健康对照组,留取体检时血样。采用荧光定量PCR法测定全血Lnc RNA SNHG17、miR-375表达水平,双荧光素酶报告实验测定LncRNA SNHG17与miR-375相互作用。分析结直肠癌患者Lnc RNA SNHG17与miR-375表达水平的关系,比较不同临床特征患者术前Lnc RNA SNHG17、miR-375表达水平,同时分析术后表达水平对结直肠癌患者预后的预测效能,比较高、低表达患者生存率。结果治疗前,结直肠癌患者LncRNA SNHG17表达水平高于健康对照者(P<0.05),miR-375表达水平低于健康对照者(P<0.05);治疗后,结直肠癌患者Lnc RNA SNHG17表达水平明显降低(P<0.05),miR-375表达水平明显升高(P<0.05)。结直肠癌患者治疗前、后LncRNA SNHG17与miR-375表达水平均呈负相关(r=-0.448、-0.603,均P<0.05);双荧光素酶报告实验证实,结直肠癌患者LncRNA SNHG17与miR-375存在相互作用(P<0.05)。不同性别、年龄、肿瘤位置、肿瘤大小、分化程度患者术前LncRNA SNHG17、miR-375表达水平比较,差异均无统计学意义(均P>0.05);不同Dukes分期患者术前Lnc RNA SNHG17、miR-375表达水平比较,差异均有统计学意义(均P<0.05)。术后LncRNA SNHG17、miR-375表达水平对复发或转移、死亡均有预测价值(均P<0.05)。LncRNA SNHG17高表达组生存率低于Lnc RNA SNHG17低表达组(P<0.05),miR-375高表达组生存率高于miR-375低表达组(P<0.05)。结论在结直肠癌发病进程中,LncRNA SNHG17和miR-375可能介导重要机制,实时监测两者对病情诊断和预后评估有帮助。Objective To analyze the expression and significance of circulating long non-coding RNA SNHG17(LncRNA SNHG17)and microRNA-375(miR375)in colorectal cancer.Methods Totally 68 patients with colorectal cancer were enrolled in the study.The expression levels of LncRNA SNHG17 and miR-375 in the blood were determined by fluorescence quantitative PCR.The interaction between LncRNA SNHG17 and miR-375 was determined by dual luciferase reporting assay.The correlation between LncRNA SNHG17 and miR-375 was analyzed.The preoperative expression levels of LncRNA SNHG17 and miR-375 in patients with different clinical characteristics were compared,and the prediction value of the preoperative expression level of LncRNA SNHG17 and miR-375 for the prognosis were analyzed.The survival rates in patients with different level of LncRNA SNHG17 and miR-375 were compared too.Results Before treatment,the expression level of circulating LncRNA SNHG17 in colorectal cancer patients was higher than that in the control group,and the expression level of miR-375 was lower than that in the control group(P<0.05).After treatment,the expression level of circulating LncRNA SNHG17 in colorectal cancer patients was decreased,while the expression level of miR-375 was increased(P<0.05).There was a linear negative correlation between the expression of LncRNA SNHG17 and miR-375 in colorectal cancer patients before and after treatment(r=-0.448,-0.603,P<0.05),and the double luciferase assay confirmed the interaction between LncRNA SNHG17 and miR-375.The difference of preoperative expression levels of LncRNA SNHG17 and miR-375 in patients with different gender,age,tumor location,tumor size and differentiation were not statistically significant(all P>0.05).The differences in preoperative expression levels of LncRNA SNHG17 and miR-375 in patients with different Dukes stages were statistically significant(all P<0.05).The survival rate of patients with highly-expressed LncRNA SNHG17 was lower than those with lowly-expressed LncRNA SNHG17 group(P<0.05),and the surviv
