摘 要:目的通过响应面法优化多序岩黄芪根中总皂苷的提取工艺并对其抗氧化活性进行研究。方法采用乙醇超声提取法对多序岩黄芪根中的总皂苷进行提取,用正丁醇萃取法分离皂苷。以总皂苷得率为考察指标,在单因素实验基础上,研究提取时间、乙醇体积分数、料液比对多序岩黄芪根中总皂苷提取的影响,并采用响应面优化多序岩黄芪根总皂苷的提取工艺。通过测定清除1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基的能力判断多序岩黄芪皂苷的抗氧化能力,并与VC进行比较。结果优化后的提取工艺为75%乙醇(V/V)、提取时间79 min、料液比1:24(m:V),此条件下5 g原料可提取到12.45 mg多序岩黄芪根皂苷;抗氧化实验结果表明:多序岩黄芪皂苷对DPPH自由基有一定的清除能力,但弱于VC。结论在最优条件下,多序岩黄芪根中总皂苷的提取率得到了提高。Objective To optimize the extraction process of total saponins from the root of Hedysarum polybotrys by response surface method and study its antioxidation activity.Methods Thesaponins were extracted by ultrasonic extraction of ethanol,and separated by n-butanol extraction.Taking the total saponin yield as an index,the effects of extraction time,ethanol volume fraction,and material ratio on the extraction of total saponinsfrom the root of Hedysarum polybotrys were studied on the basis of single factor experiments,and theextraction process of total saponin was optimized by response surface method.The antioxidant capacity of astragalosides was determined by the ability of scavenging 1,1-diphenyl-2-picrylhydrazyl(DPPH) free radicals,and compared with VC.Results The optimum extraction process were 75% ethanol(V/V),the extraction time 79 min and the solid-liquid ratio 1:24(m:V),under this condition,12.45 mg astragalosidecould be extracted from 5 g materials.The results of antioxidant experiment showed that astragaloside from polysucophane had a certain scavenging ability to DPPH free radicals,but weaker than VC.Conclusions Under the optimal conditions,the extraction rate of total saponins from the root of Hedysarumpolybotrys is improved.
