摘  要:目的探讨白血病患儿与健康儿童血液中磷脂酰肌醇激酶(PI3K)、蛋白激酶B (Akt)、乳酸脱氢酶(LDH)因子含量的变化。方法采用Western blot方法,检测骨髓核细胞中PI3K、Akt、p53、Caspase-3蛋白水平;采用RT-PCR方法,检测PI3K/Akt的mRNA表达;直接取血清测量乳酸脱氢酶(LDH)含量的变化。结果初治急性白血病患儿与复发性白血病患儿骨髓核细胞中PI3K、Akt、p53蛋白含量、PI3K mRNA、Akt mRNA水平、外周血清LDH含量明显高于健康儿童,而Caspase-3蛋白水平明显减少(P<0.05);初治急性白血病患儿骨髓核细胞中Akt蛋白、Akt mRNA水平高于复发性白血病患儿(P<0.05),而PI3K蛋白、PI3K mRNA含量低于复发性白血病患儿(P<0.05)。结论可通过调控PI3K/Akt等因子变化来影响白血病细胞增殖与凋亡。Objective To study the changes of phosphatidylinositol 3-kinase(PI3K),protein kinase B(Akt)and lactate dehydrogenase(LDH)in the blood of children with leukemia and healthy children.Methods Western blot was used to detect the protein levels of PI3K,Akt,p53 and Caspase-3 protein in bone marrow nucleus cells.RT-PCR was used to detect the mRNA expression of PI3K/Akt.The serum was taken directly to measure the change of lactate dehydrogenase(LDH).Results The levels of PI3K,Akt and p53 proteins,PI3K mRNA and Akt mRNA,peripheral serum LDH in bone marrow nucleus cells of children with newly-treated acute leukemia and children with relapsed leukemia were significantly higher than those of healthy children,while the protein level Caspase-3 was significantly reduced(P<0.05).The levels of Akt protein and Akt mRNA in bone marrow nucleus cells of patients with newly-treated acute leukemia were higher than those of children patients with relapsed leukemia(P<0.05),while the levels of PI3K protein and PI3K mRNA were lower than those of children patients with relapsed leukemia(P<0.05).Conclusion The regulation of changes in factors such as PI3K/Akt can affect the proliferation and apoptosis of leukemia cells.
