摘 要:探讨人GRP94(葡萄糖调节蛋白94)在宫颈癌组织中的表达及其对宫颈癌细胞生物学行为的影响。收集2015年1月至2016年7月成都医学院第一附属医院有完整病历资料的46例宫颈癌患者组织标本及对应癌旁组织,实时荧光定量-聚合酶链反应(qRT-PCR)检测宫颈癌组织中GRP94表达水平;采用脂质体转染法将GRP94表达质粒转染入宫颈癌Hela细胞,上调细胞内源GRP94表达;活细胞计数试剂盒(CCK-8)和流式细胞术检测GRP94对宫颈癌Hela细胞增殖及凋亡的影响;Transwell小室法检测其对Hela细胞迁移及侵袭能力的影响。46例宫颈癌患者癌组织中GRP94表达量高于癌旁组织,差异有统计学意义(P<0.05);进一步分析宫颈癌患者癌组织中GRP94表达与患者临床指标发现,GRP94的表达水平与患者的年龄无关(P>0.05),而与FIGO分期以及病例分级密切相关(P<0.05);转染GRP94表达质粒后,上调宫颈癌Hela细胞内源GRP94表达;相比于对照组和空白组,GRP94转染组的宫颈癌细胞增殖能力显著升高(P<0.05),凋亡率下降(P<0.05),细胞的侵袭和迁移能力均显著升高(P<0.05)。结果显示GRP94在宫颈癌组织中的表达量显著升高,其表达水平的升高与宫颈癌的FIGO分期以及分化程度密切相关;在宫颈癌细胞中的上调GRP94的表达,能够明显提高其细胞增殖、迁移和侵袭,抑制细胞凋亡,表明GRP94的表达与宫颈癌发生发展密切相关。To investigate the expression of human GRP94(glucose regulatory protein 94)in cervical cancer tissues and its effect on the biological behavior of cervical cancer cells,collecting 46 specimens of cervical cancer patients with complete medical records from January 2015 to July 2016 from the First Affiliated Hospital of Chengdu Medical College,Department of Gynecology.Then corresponding adjacent tissues,real-time fluorescence quantitative-polymerase chain reaction(qRT-PCR)for detecting GRP94 expression in cervical cancer tissues Level;Transfection of GRP94 expression plasmid into cervical cancer Hela cells by lipofection,up-regulation of endogenous GRP94 expression;Viable cell counting kit(CCK-8)and flow cytometry for detection of GRP94 on cervical cancer Hela cells effects of proliferation and apoptosis;Transwell chamber assay for its effect on Hela cell migration and invasion was observed.The expression of GRP94 in 46 cases of cervical cancer patients was higher than that in adjacent tissues(P<0.05).Further analysis of GRP94 expression in cervical cancer patients and clinical indicators showed that GRP94 expression level and patients age was not related(P>0.05),but closely related to FIGO stage and case classification(P<0.05).Transfection of GRP94 expression plasmid up-regulated the expression of endogenous GRP94 in cervical cancer Hela cells;Compared with control group and blank group,the proliferation of cervical cancer cells in GRP94 transfection group was significantly increased(P<0.05),the apoptosis rate decreased(P<0.05),and the invasion and migration ability of cells were significantly increased(P<0.05).The expression of GRP94 in cervical cancer tissues is significantly increased.The increase of expression level is closely related to the FIGO stage and degree of differentiation of cervical cancer.Up-regulation of GRP94 expression in cervical cancer cells can significantly increase cell proliferation and migration.And invasion,and inhibition of apoptosis,are indicating that the expression of GRP94 is closel
