幽门螺杆菌感染病人胃癌组织中MMP-7、VEGF的表达及相关性研究

摘　　要：目的:检测幽门螺杆菌(Hp)感染与基质金属蛋白酶-7(MMP-7)、血管内皮生长因子(VEGF)在胃癌组织中的表达及临床意义。方法:胃癌组织来源于2015年1月至2018年11月蚌埠医学院第一附属医院普外科手术室、肿瘤外科手术室,按照幽门螺杆菌是否感染分2组。采用荧光定量PCR法及免疫组织化学检测MMP-7、VEGF的表达。结果:60例胃癌组织中,Hp阳性组、有浆膜层浸润、有淋巴结转移的胃癌病人MMP-7、VEGF mRNA表达高于Hp阴性组、无浆膜层浸润、无淋巴结转移的病人(P<0.01);MMP-7与VEGF mRNA表达值之间呈正相关(r=0.498,P<0.01);Hp与MMP-7及VEGF表达呈正相关,MMP-7和VEGF的表达呈正相关(P<0.05~P<0.01)。结论:MMP-7、VEGF与胃癌的发生、发展可能有关,在胃癌的发生、发展过程中幽门螺杆菌与MMP-7、VEGF可能起到了相互协同的作用。MMP-7与VEGF表达呈正相关。Objective:To detect the expression levels and clinical significance of matrix metalloproteinase-7(MMP-7)and vascular endothelial growth factor(VEGF)in gastric cancer tissues of patients with Helicobacter pylori(Hp)infection.Methods:The gastric cancer tissues were collected from Department of General Surgery and Tumor Surgery Operating Rooms of the First Affiliated Hospital of Bengbu Medical College from January 2015 to November 2018,and were divided into two groups according to whether with Hp infection.The expression levels of MMP-7 and VEGF were detected using the fluorescence quantitative PCR and immunohistochemistry.Results:Among 60 cases of gastric cancer tissues,the mRNA expression levels of MMP-7 and VEGF in Hp-positive group,patients with serous layer infiltration and lymph node metastasis were higher than in Hp-negative group,patients without serous layer infiltration and lymph node metastasis(P<0.01).There was a positive correlation between MMP-7 and VEGF mRNA expressions(r=0.498,P<0.01).The Hp was positively correlated with MMP-7 and VEGF expression levels,and the expression level of MMP-7 was positively correlated with VEGF(P<0.05 to P<0.01).Conclusions:MMP-7 and VEGF may be related to the occurrence and development of gastric cancer,and Hp,MMP-7 and VEGF may play a synergistic role in the occurrence and development of gastric cancer.The expression level of MMP-7 is positively correlated with VEGF.
