摘  要:目的研究大蒜素对心肌缺血大鼠心肌组织降钙素基因相关肽(CGRP)及其受体mRNA表达水平的影响,旨在探讨心肌缺血的发病机制及大蒜素的作用机制。方法腹腔注射异丙肾上腺素建立大鼠心肌缺血模型,将建模成功的大鼠随机分为模型组、大蒜素高剂量组、大蒜素中剂量组、大蒜素低剂量组,另选取正常大鼠作为正常对照组,每组12只。大蒜素各剂量组分别灌胃大豆油配制的高、中、低浓度的大蒜素,模型组和正常对照组灌胃等容量的大豆油,均灌胃30 d。酶联免疫吸附法测定各组大鼠心肌组织CGRP水平,实时荧光定量聚合酶链式反应检测CGRP受体mRNA的表达情况。结果与正常对照组比较,大蒜素各剂量组和模型组心肌组织CGRP受体mRNA表达水平均降低(P<0.05)。与模型组比较,大蒜素低、中剂量组心肌组织CGRP受体mRNA表达水平均增高(P<0.05)。结论心肌缺血大鼠心肌组织CGRP受体mRNA表达水平降低,一定剂量的大蒜素可使大鼠心肌组织CGRP受体mRNA表达水平升高,增强心肌组织中CGRP的扩血管作用,缓解心肌缺血症状,对心肌具有保护作用。Objective To investigate the effects of allicin on the expression of calcitonin gene-related peptide(CGRP)receptor mRNA in myocardial tissue of rats with myocardial ischemia.Methods The rat models of myocardial ischemia were established with intraperitoneal injection of isoproterenol.The myocardial ischemia rats were randomly divided into low-dose group,middle-dose group,high-dose group,and model group,with 12 rats in each group,another 12 normal rats were taken as the normal control group.The rats in each dose group were intragastrically administered with different doses of allicin for 30 days.The rats in model group and normal control group were given the same amount of soybean oil for 30 days.The levels of CGRP in the myocardial tissue were determined by enzyme linked immunosorbent assay.The levels of CGRP mRNA in the myocardial tissue of each group were determined by real-time fluorescence quantification polymerase chain reaction.Results Compared with control group,the expression levels of CGRP mRNA in myocardial tissue of each dose group and model group were significantly decreased(P<0.05).Compared with model group,the expression levels of CGRP mRNA in myocardial tissue of low dose group and middle dose group were significantly increased(P<0.05).Conclusion The expression levels of CGRP mRNA in myocardial tissue of rats with myocardial ischemia were decreased.A certain dose of allicin could increase the expression of CGRP mRNA in rat myocardial tissue,enhance the effect of CGRP vasodilation in myocardial tissue,relieve the symptoms of myocardial ischemia,and has a protective effect on myocardium.
