摘  要:目的 探讨CO2气腹对裸鼠卵巢癌移植瘤中尿激酶型纤溶酶原激活物(uPA)、尿激酶型纤溶酶原激活物受体(uPAR)、尿激酶型纤溶酶原激活抑制物(PAI-1)表达的影响.方法 40只雌性Balb/c裸鼠腹腔注射卵巢癌细胞株SKOV3构建卵巢癌移植瘤模型,随机分为对照组、开腹组、CO2气腹0.5 h组、CO2气腹1h组;造模后12周处死并取瘤体组织,采用免疫组化、实时荧光定量PCR与Western blot检测移植瘤中uPA、uPAR、PAI-1表达.结果 CO2气腹组移植瘤中uPA、uPAR、PAI表达明显高于对照组、开腹组(P<0.05),但CO2气腹0.5 h、1h组差异无统计学意义(P>0.05).结论 CO2气腹可能通过上调uPA、uPAR、PAI表达促进卵巢癌生长和转移.Objective To investigate the effect of carbon dioxide(CO2)pneumoperitoneum on urokinase type plasminogen activator(uPA),urokinase type plasminogen activator receptor(uPAR)and urokinase type plasminogen activator inhibitor-1(PAI-1)in nude mice with ovarian cancer xenograft.Methods Forty female nude mice were intraperitoneally injected with ovarian cancer cell line SKOV3 to establish the ovarian cancer xenograft model and then randomized to control,laparotomy,and CO2 pneumoperitoneum(0.5 h,1 h)groups.Expression of uPA,uPAR and PAI in xenograft was detected by immunohistochemistry,quantitative real-time PCR and Western blot after 12 weeks.Results Expression of uPA,uPAR and PAI was higher in CO2 pneumoperitoneum groups than in control and laparotomy groups(P<0.05),while their expression was comparable between CO2 pneumoperitoneum groups(P>0.05).Conclusion CO2 pneumoperitoneum may promote the growth and metastasis of ovarian cancer via up-regulating expression of uPA,uPAR and PAI.
