胃癌组织中LETM1、HIF-1α的表达变化及意义

摘　　要：目的观察胃癌组织中亮氨酸拉链EFhand结构域跨膜蛋白1(LETM1)与缺氧诱导因子-1α(HIF-1α)的表达变化,并探讨其意义。方法选取胃癌患者95例,应用免疫组化技术检测胃癌组织及癌旁正常组织中LETM1和HIF-1α表达,并分析二者表达与患者临床病理特征的关系。随访5年,采用Kaplan-Meier法分析LETM1、HIF-1α表达与患者预后的关系。结果胃癌组织中LETM1和HIF-1α阳性表达率高于癌旁正常组织(P均<0.05)。胃癌组织中LETM1和HIF-1α表达与浸润深度、临床TNM分期、组织分化程度及淋巴结转移有关(P均<0.05),与患者性别、年龄、组织学分型及肿瘤直径无关(P均>0.05)。相关分析结果显示,胃癌患者LETM1和HIF-1α表达呈正相关(r=0.362,P<0.05)。Kaplan-Meier分析显示,LETM1和HIF-1α检出结果阴性的患者5年生存率高于结果阳性的患者(P均<0.05)。结论胃癌组织中LETM1和HIF-1α表达均升高,二者协同促进胃癌的发生发展;检测二者表达有助于胃癌诊断及病情判断。Objective To observe the expression changes of leucine zipper-EFhand domain transmembrane protein 1(LETM1)and hypoxia-inducible factor-1α(HIF-1α)in the gastric cancer tissues and to explore its clinical significance.Methods Ninety-Five patients with gastric cancer were selected.Immunohistochemistry was used to analyze the expression of LETM1 and HIF-1αin the gastric cancer tissues and adjacent normal tissues.The relationship between the expression of these two indexes and clinicopathological features was analyzed.The follow-up time was 5 years.Kaplan-Meier test was used to analyze the relationship between LETM1 and HIF-1αexpression and patients′prognosis.Results The positive expression rates of LETM1 and HIF-1αin the gastric cancer tissues were significantly higher than those in the adjacent tissues,and the difference was statistically significant(both P<0.05).The expression levels of LETM1 and HIF-1αin the gastric cancer tissues were related to depth of invasion,clinical TNM stage,differentiation and lymph node metastasis(all P<0.05),not to gender,age,tumor histology type or tumor diameter(all P>0.05).Correlation analysis showed that the expression of LETM1 and HIF-1αin the gastric cancer patients was positively correlated(r=0.362,P<0.05).Kaplan-Meier analysis showed that the 5-year survival rate of patients with negative detection of LETM1 and HIF-1αwas significantly higher than that of patients with positive results(P<0.05).Conclusion The expression levels of LETM1 and HIF-1αin the gastric cancer tissues increase,which may synergistically promote the occurrence and development of gastric cancer;detection of both expression is helpful for the diagnosis and judgement of gastric cancer.
