摘 要:目的探讨miR-621在结直肠癌组织中的表达及对细胞活性、细胞增殖和迁移侵袭的影响。方法选择20对结直肠癌患者的癌组织及其相应癌旁正常组织(距癌组织边缘>5 mm),采用qRT-PCR法检测miR-621在人结直肠癌组织和癌旁正常组织中的表达。合成可抑制miR-621表达的甲氧基修饰的反义寡核苷酸链ASO-miR-621后,取对数生长期的结直肠癌细胞HCT116和SW480,分为NC组、miR-621组、ASO-NC组、ASO-miR-621组,分别转染pcDNA3、pcDNA3-miR-621、ASO-NC及ASO-miR-621,采用MTT法、细胞集落形成实验及Transwell迁移侵袭实验研究miR-621在结直肠癌细胞中的作用。结果miR-621在结直肠癌组织中表达低于其相应癌旁正常组织(P<0.05);miR-621组细胞生长活性、增殖和迁移侵袭能力低于NC组(P均<0.05),ASO-miR-621组细胞生长活性、增殖和迁移侵袭能力高于ASO-NC组(P均<0.05)。结论miR-621在结直肠癌组织中呈低表达;miR-621可抑制结直肠癌细胞的活性、增殖和迁移侵袭,起到抑癌基因的作用。Objective To investigate the expression of miR-621 in the colorectal cancer tissues and its effects on cell activity,cell proliferation,migration,and invasion.Methods Twenty pairs of cancer tissues and normal tissues adjacent to cancer(>5 mm from cancer tissues)from patients with colorectal cancer were selected.qRT-PCR was used to detect the expression of miR-621 in human colorectal cancer tissues and its corresponding normal tissues.After synthesizing the methoxy-modified antisense oligonucleotide chain ASO-miR-621 which can inhibit the expression of miR-621,we chose colorectal cancer cells HCT116 and SW480 in the logarithmic phase and divided them into the NC group,miR-621 group,ASO-NC group,and ASO-miR-621 group,which were transfected with pcDNA3,pcDNA3-miR-621,ASO-NC,and ASO-miR-621,respectively.MTT method,cell colony formation experiment and Transwell migration and invasion experiment were used to study the role of miR-621 in the colorectal cancer cells.Results MiR-621 expression in the colorectal cancer tissues was lower than that of its corresponding normal tissues(P<0.05);the miR-621 group had lower cell growth activity,proliferation,migration and invasion abilities than the NC group(all P<0.05).The cell growth activity,proliferation,migration and invasion abilities of the ASO-miR-621 group were higher than those of the ASO-NC group(all P<0.05).Conclusion MiR-621 is low expressed in the colorectal cancer tissues,which can inhibit the activity,proliferation,migration and invasion of colorectal cancer cells and play a role as a tumor suppressor gene.
