摘 要:目的分析鼻咽癌癌组织中CD133、丝裂原活化蛋白激酶4(MKK4)、需肌醇酶1(p-IRE1)蛋白表达与放疗预后的相关性,以期为临床诊治鼻咽癌及评估预后提供信息。方法将2012年5月-2013年12月于本院开展放疗治疗的94例鼻咽癌患者作为观察对象,经SP免疫组化法对鼻咽癌患者癌组织内CD133、MKK4、p-IRE1蛋白表达情况进行分析,并给予患者放射治疗;探讨癌组织中CD133、MKK4、p-IRE1蛋白表达和鼻咽癌临床特征、放疗预后相关性。结果鼻咽癌癌组织内CD133、MKK4、p-IRE1蛋白的阳性表达率分别为68. 1%、75. 5%、67. 0%;35例患者呈CD133低表达,59例患者呈CD133高表达;38例患者呈MKK4低表达,56例患者呈MKK4高表达;69例患者呈p-IRE1低表达,25例患者呈p-IRE1高表达;Logistic回归分析发现:癌组织内CD133、MKK4、p-IRE1蛋白阳性表达与鼻咽癌临床TNM分期、淋巴结转移存在相关性(P <0. 05);鼻咽癌癌组织内CD133、MKK4、p-IRE1蛋白低表达患者放疗后5年总生存率均高于CD133、MKK4、p-IRE1蛋白高表达患者(P <0. 05)。结论 CD133、MKK4蛋白于鼻咽癌的癌组织内均呈高表达,p-IRE1蛋白于鼻咽癌的癌组织内呈低表达;同时,癌组织内CD133、MKK4、p-IRE1蛋白表达和鼻咽癌临床分期、淋巴结转移及放疗预后关系密切,可作临床诊治鼻咽癌及评估预后的重要指标。Objective To analyze the correlation between the expressions of CD133,mitogen-activated protein kinase 4(MKK4),inositolase 1(p-IRE1)protein and the prognosis of nasopharyngeal carcinoma,in order to provide information for the diagnosis and treatment of clinical nasopharyngeal carcinoma and evaluate the prognosis of patients.Methods A total of 94 patients with nasopharyngeal carcinoma who underwent radiotherapy in our hospital from May 2012 to December 2013 were included as subjects.The expressions of CD133,MKK4 and p-IRE1 protein in cancer tissues of patients with nasopharyngeal carcinoma were analyzed by SP immunohistochemistry.The relationship between CD133,MKK4,p-IRE1 protein expression and clinical features of nasopharyngeal carcinoma and the prognosis of radiotherapy was investigated.Results The positive expression rates of CD133,MKK4 and p-IRE1 in nasopharyngeal carcinoma were 68.1%,75.5%and 67.0%,respectively.35 patients showed low expression of CD133,59 patients showed high expression of CD133;38 patients showed low expression of MKK4,and 56 patients showed high expression of MKK4;69 patients showed low expression of p-IRE1,and 25 patients showed high expression of p-IRE1;Logistic regression analysis showed that there was a correlation between the positive expressions of CD133,MKK4 and p-IRE1 protein and the clinical TNM stage and lymph node metastasis of nasopharyngeal carcinoma(P<0.05).The 5-year overall survival rate of patients with low expressions of CD133,MKK4 and p-IRE1 in nasopharyngeal carcinoma was higher than that of CD133,MKK4 and p-IRE1 protein(P<0.05).Conclusion CD133 and MKK4 proteins are highly expressed in the tissues of nasopharyngeal carcinoma,and p-IRE1 protein is low in the tissues of nasopharyngeal carcinoma.At the same time,the expressions of CD133,MKK4 and p-IRE1 protein in cancer tissues are closely related to the clinical stage,lymph node metastasis and prognosis of nasopharyngeal carcinoma,which can be used as an important index for the clinical diagnosis,treatment and prognosis
