The eukaryotic transcription factor NF-κB is involved in the regulation of inflammation, immune response, programmed cell death, cell proliferation and differentiation by regulating the expression of multiple target genes. RelA is one of the most important members of the NF-κB family. Its post-translational modification can precisely regulate the transcriptional activation of NF-κB, and plays an important role in inflammation, tumor, metabolism, immune, and the occurrence and development of related diseases. In this review, we summarized the types, regulatory mechanisms, effects of post-translational modifications of RelA, and discussed its functions in various diseases such as NF-κB-mediated inflammation and cancer.
