摘  要:据报道,有超过50%的miRNA位于或靠近基因组中肿瘤相关的脆性区域,即经常发生缺失、扩增、易位的染色体片段,并且与人类肿瘤的发生及发展密切相关。MiRNA通过与靶mRNA的3’-未翻译区域内的互补结合位点结合,抑制或促进mRNA的翻译或降解。MiR-449已经被检测出有多种靶标,并且可以通过调控基因表达来作用于细胞发育、细胞分化、凋亡、增殖、应激反应和脂肪代谢。MiR-449家族的异常表达在多种癌症疾病中得到证实,如肝癌、胃癌、肺癌、乳腺癌、结直肠癌等。基于肿瘤与miR-449之间可能的致病关系,其有可能作为治疗某些类型癌症的潜在药物。本文综述了miR-449家族在癌症中的生物学功能和相关的分子机制,以期为miR-449作为潜在的癌症治疗药物提供证据。According to reports,more than 50%of miRNAs are located in or close to tumor-associated fragile regions in the genome,which are chromosomal fragments that been frequently deleted,amplified or translocated,and are closely related to the occurrence and development of human tumors.MiRNAs bind to complementary binding sites in the 3'-untranslated region of the target mRNA to inhibit or promote translation or degradation of the mRNAs.MiR-449 has been detected with multiple targets and work on cell development,cell differentiation,apoptosis,proliferation,stress response and fat metabolism by regulating gene expression.Abnormal expression of the miR-449 family has been confirmed in various cancer diseases,such as liver cancer,gastric cancer,lung cancer,breast cancer,and colorectal cancer.On the basis of the putative pathogenetic relationships between cancer and miR-449,it may be a potential way for the treatment of certain types of cancer.This review summarized the biological functions of the miR-449 family in cancer and the related molecular mechanisms,in order to provide evidence for miR-449 as a potential cancer treatment method.
