摘  要:目的探讨乳铁蛋白对神经病理性疼痛大鼠的镇痛作用及对炎性细胞因子的影响。方法通过坐骨神经慢性压迫性损伤(CCI)诱导神经病理性疼痛模型。将40只模型大鼠随机分为A、B、C、D和E组,每组8只。建模后第5天开始给药。A组给予0.9%NaCl 1 mL;B,C,D组分别给予0.5,1.0和2.0 g·kg^-1乳铁蛋白;E组给予1 g·kg^-1乳铁蛋白和2 mg·kg^-1纳洛酮。5组大鼠每天腹腔注射给药1次,连续5 d。比较5组大鼠行为学[机械缩足反射阈值(MWT)和热缩足反射潜伏期(TWL)]和炎性细胞因子水平变化[肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)]。结果 5组大鼠均造模成功。术后第9天后,A,B,C,D和E组的MWT分别为(8.87±0.61),(16.08±0.44),(17.32±0.52),(16.77±0.31)和(16.23±0.63)g,TWL分别为(7.43±0.49),(11.94±0.45),(12.22±0.31),(11.99±0.27)和(11.32±0.36)s,TNF-α分别为(121.11±9.62),(86.73±9.26),(79.47±9.67),(81.62±10.13)和(89.03±9.73)ng·L^-1,IL-6分别为(33.98±5.22),(26.26±4.61),(25.57±4.72),(26.36±4.26)和(26.65±4.36)ng·L^-1。C组的上述指标与A组和E组比较,差异均有统计学意义(均P<0.05)。结论乳铁蛋白可以缓解神经病理性疼痛大鼠的痛觉异常,并减轻炎症反应。Objective To investigate the effect of lactoferrin on pain threshold and inflammatory cytokines in rats with neuropathic pain.Methods The neuropathic pain rats were established by chronic constriction injury(CCI).Forty SD rats were randomly divided into groups A(0.9%NaCl 1 mL),B,C,D(0.5,1.0 and 2.0 g·kg^-1 lactoferrin)and E(1.0 g·kg^-1 lactoferrin+2 mg·kg^-1 naloxone),8 rats in each group.The drug was administered on the 5th day after successful modeling.All groups received intraperitoneal injection once a day for 5 days.The behavioral changes[mechanical withdrawal threshold(MWT)and thermal withdrawal latency(TWL)]and inflammatory cytokin levels[interleukin-6(IL-6)andtumor necrosis factor-α(TNF-α)]were compared among five groups.Results All the rats in the 5 groups were successfully established.Nine days after operation,the main indexes of groups A,B,C,D and E were compared:MWT were(8.87±0.61),(16.08±0.44),(17.32±0.52),(16.77±0.31)and(16.23±0.63)g,TWL were(7.43±0.49),(11.94±0.45),(12.22±0.31),(11.99±0.27)and(11.32±0.36)s,TNF-αwere(121.11±9.62),(86.73±9.26),(79.47±9.67),(81.62±10.13)and(89.03±9.73)ng·L^-1,IL-6 were(33.98±5.22),(26.26±4.61),(25.57±4.72),(26.36±4.26)and(26.65±4.36)ng·L^-1.The above-mentioned indexes in group C were significantly different from those in groups A and E(all P<0.05).Conclusion Lactoferrin can relieve the abnormal pain sensation and reduce the inflammatory response in rats with neuropathic pain.
