摘 要:目的研究β-榄香烯对人卵巢癌细胞Cavo-3增殖的影响及机制探索。方法体外培养卵巢癌Cavo-3细胞,用MTT实验和集落形成实验检测β-榄香烯对卵巢癌Cavo-3细胞增殖的影响,Western blot实验检测β-榄香烯对Cavo-3细胞EGFR及其下游信号通路的影响。结果β-榄香烯以浓度和时间依赖性方式抑制卵巢癌Cavo-3细胞的增殖;Western blot实验发现β-榄香烯能显著抑制EGFR及其下游通路磷酸化水平,而不影响总蛋白水平。结论β-榄香烯通过EGFR信号通路抑制卵巢癌Cavo-3细胞的增殖,揭示了β-榄香烯发挥抗卵巢癌作用的新机制。Objective To investigate the effect and mechanism ofβ-elemene on the proliferation of ovarian cancer cavo-3 cells.Methods Ovarian cancer cavo-3 cells were cultured in vitro,effect ofβ-elemene on the proliferation of ovarian cancer cavo-3 cells was determined by MTT and colony formation assay,at the same time Western blot assay was used to detect the effect ofβ-elemene on EGFR and its downstream signaling pathway in ovarian cancer cavo-3 cells.Resultsβ-elemene inhibited the proliferation of ovarian cancer cavo-3 cells in a concentration-dependent and timedependent manner,western blot analysis showed thatβ-elemene significantly inhibited the phosphorylation of EGFR and its downstream pathway,without affecting the total protein level.Conclusionβ-elemene inhibits the proliferation of ovarian cancer cavo-3 cells through the EGFR signaling pathway and plays an anti-tumor role.
