摘　　要：[目的]探讨YAP在大肠癌中的表达情况,及其对大肠癌细胞增殖和侵袭能力的影响。[方法]运用生物信息学方法分析YAP在大肠癌组织和正常大肠组织中的表达情况;应用Western blot方法检测大肠癌及其癌旁组织中YAP的表达情况;建立干扰YAP表达的大肠癌细胞株HCT116和SW480,通过MTT法和Transwell实验检测YAP对大肠癌细胞增殖和侵袭能力的影响。[结果]对筛选出的GSE9348和GSE5206两组芯片数据分析均提示YAP mRNA在大肠癌组织中的表达水平明显高于正常大肠组织(P<0.01);Western blot实验结果显示大肠癌组织中YAP的表达水平明显高于癌旁组织(P<0.01);MTT法动态观察发现敲低YAP基因后大肠癌细胞HCT116和SW480的增殖受到明显的抑制(P<0.01);Transwell小室实验提示敲低YAP基因后大肠癌细胞跨膜细胞数明显减少(P<0.001)。[结论]YAP在大肠癌中高表达,YAP促进大肠癌细胞的增殖和侵袭,可能是大肠癌治疗的潜在靶点。[Objective]To investigate the expression of Yes-associated protein(YAP)in colorectal cancer and effect of YAP on the proliferation and invasion of colorectal cancer.[Methods]The difference expression of YAP between colorectal cancer and normal colorectal tissue was studied through bioinformatics analysis.Western blot was used to detect YAP expression in colorectal cancer and normal colorectal tissue.The colorectal cancer cell lines(HCT116and SW480)with YAP knockdown were constructed;cell proliferation was measured by MTT assay;cell invasion ability was tested through transwell invasion assay.[Results]GSE9348and GSE20842two group of microarray data showed that the expression level of YAP mRNA in colorectal cancer was significantly higher than normal colorectal tissue(P<0.01).Western blot test results showed that the expression level of YAP protein in colorectal cancer tissues was higher than that in its adjacent normal colorectal tissues(P<0.01).MTT dynamic observation showed that the proliferation of colorectal cancer cell lines with YAP knockdown was significantly inhibited compared with the control group(P<0.01).The results from transwell assay revealed that the migration cell numbers of colorectal cancer cells with YAP knockdown decreased markedly compared with the control group(P<0.01).[Conclusions]The expression of YAP is upregulated in colorectal cancer.YAP can promote the proliferation and invasion ability of colorectal cancer cells.It may be a potential target for treatment of colorectal cancer.
