摘　　要：目的检测RasGRP3蛋白在肝癌组织及其癌旁组织中的表达水平,结合临床病理资料,探讨RasGRP3蛋白与肝癌患者临床病理特征的关系。方法回顾性分析2000年12月至2015年12月安徽医科大学第二附属医院100例HCC患者手术标本及其临床资料(作者贺良为安医大二附院普外科医生,协同完成,提供病例资料及标本),采用组织免疫组化方法分析肝癌组织及其癌旁组织中的RasGRP3蛋白表达情况,利用 2检验及Fisher确切概率法分析肿瘤组织及癌旁组织蛋白表达程度及其表达程度与肿瘤临床病理分期,TNM分期,肿瘤体积及预后生存时间的关系;Kaplan-Meier分析肝癌患者生存数据与RasGRP3蛋白表达的相关性,绘制生存曲线,log-rank cox用于检验差异性。结果肿瘤组织中RasGRP3蛋白表达58例(58%),癌旁组织中RasGRP3蛋白表达42例(42%);在肝癌组织和癌旁组织中,RasGRP3蛋白的表达程度差异具有统计学意义(P<0.05),RasGRP3蛋白在肝癌组织中的表达与肿瘤的临床病理分期(P<0.01)、TNM分期(P<0.01)、肿瘤体积(P<0.05)、预后生存明显相关(P<0.01)。结论 RasGRP3蛋白在肝癌中表达增高,且其表达在肝癌发生发展中发挥重要作用,为肝癌分子靶向治疗提供新思路。Objective To detect the expression level of RasGRP3 protein in hepatocarcinoma tissues and adjacent tissues,and to analyze the relationship between RasGRP3 protein and clinicopathological features of liver cancer patients.Methods A retrospective analysis of 100 cases of HCC patients with surgical specimens and clinical data from the Second Affiliated Hospital of Anhui Medical University from December 2000 to December 2015,tissue immunohistochemistry was used to analyze the expression of RasGRP3 protein in hepatocarcinoma tissues and adjacent tissues,and the expression of tumor tissue and adjacent tissues was analyzed by 2 test and Fisher exact probability method.Pathological staging,TNM staging,tumor volume and prognosis survival time;Kaplan-Meier analysis of the correlation between survival data of liver cancer patients and RasGRP3 protein expression,mapping survival curve log-rank cox for testing differential log-rank cox for testing differences.Results The expression of RasGRP3 protein in tumor tissues was 58(58%),and the expression of RasGRP3 protein in paracancerous tissues was 42(42%).The expression of RasGRP3 protein in liver cancer tissues and adjacent tissues was statistically significant(P<0.05),the expression of RasGRP3 protein in hepatocarcinoma tissues was significantly correlated with clinical pathological stage(P<0.01),TNM stage(P<0.01),tumor volume(P>0.05),and prognosis survival(P<0.01).Conclusion RasGRP3 protein is highly expressed in hepatocarcinoma,and its expression plays an important role in the development of hepatocarcinoma,which provides a new idea for molecular targeted therapy of liver cancer.
