摘  要:目的考察仙鹤草不同提取物对运动性疲劳小鼠抗疲劳及耐缺氧能力的影响。方法将KM种小鼠随机分为空白组20只和造模组140只,造模组小鼠定时分批进行游泳训练2周。选造模达标运动性疲劳小鼠随机分为模型组、水提取物组、70%乙醇组、正丁醇组、石油醚组、乙酸乙酯组、二氯甲烷组,20只/组。各给药组在游泳训练后30 min灌胃给予相应药物(生药量)2.4 g/kg,空白组及模型组分别给予同体积蒸馏水,均1次/d,连续1周。末次给药30 min后,每组随机取10只小鼠,测定血清乳酸(LAC)、尿素氮(BUN)水平及肝组织中肝糖原(LG)含量和脑组织中谷氨酸(GLU)、γ-氨基丁酸(GABA)、5-羟色胺(5-HT)含量;各组剩余10只小鼠依次放入内置10 g新鲜钠石灰的广口瓶内,记录封口后至小鼠呼吸停止所需时间(耐缺氧存活时间)。结果模型组小鼠血清LAC、BUN水平均显著高于空白组(P均<0.05),肝组织中LG含量显著低于空白组(P<0.05),耐缺氧存活时间显著短于空白组(P<0.05);水提取物组、正丁醇组、乙酸乙酯组小鼠血清LAC水平和70%乙醇组、正丁醇组、乙酸乙酯组血清BUN水平均显著低于模型组(P均<0.05),水提取物组、70%乙醇组、正丁醇组、乙酸乙酯组小鼠肝组织中LG含量显著高于模型组(P均<0.05),正丁醇组、乙酸乙酯组小鼠耐缺氧存活时间显著长于模型组(P均<0.05)。模型组小鼠脑组织中GLU含量显著低于空白组(P<0.05),GABA、5-HT含量均显著高于空白组(P均<0.05);各给药组小鼠脑组织中GLU、GABA含量与模型组比较差异均无统计学意义(P均>0.05),正丁醇组小鼠脑组织中5-HT含量与模型组比较显著降低(P<0.05)。结论仙鹤草的正丁醇与乙酸乙酯提取物可能是其抗疲劳的主要有效部位,作用机制与干预机体能量代谢、保护缺氧组织有关,其正丁醇提取物对中枢疲劳也有一定影响。Objective It is to investigate the effects of different extracts of agrimony on the anti-fatigue and hypoxic-resistant ability of mice with exercise fatigue.Methods The KM mice were randomly divided into blank group(n=20)and molding group(n=140).The mice in the molding group were regularly trained in batches for 2 weeks.The exercise fatigue mice that met the molding standard were randomly divided into model group,water extract group,70%ethanol group,n-butanol group,petroleum ether group,ethyl acetate group,dichloromethane group,20 mice/group.Each medication group was given 2.4 g/kg of the corresponding drug(crude drug)by intragastric administration at 30 minutes after swimming training,the blank group and the model group were given the same volume of distilled water,all once a day,for 1 week.At 30 minutes after the last administration,10 mice were randomly selected from each group,and the levels of serum lactic acid(LAC),urea nitrogen(BUN),and the contents of liver glycogen(LG)in liver tissue and glutamate(GLU),γaminobutyric acid(GABA)and serotonin(5-HT)in brain tissue were measured.The remaining 10 mice in each group were placed in a wide-mouth bottle with 10g of fresh sodium lime in turn,and the time from sealing to stopping breathing of the mice was recorded(hypoxia-resistant survival time).Result The serum levels of LAC and BUN in the model group were significantly higher(all P<0.05),the LG content in liver tissue was significantly lower(P<0.05),and the hypoxia-resistant survival time was significantly shorter than that in the blank group Group(P<0.05);the serum LAC level of mice in water extract group,n-butanol group,ethyl acetate group and the serum BUN level of 70%ethanol group,n-butanol group,ethyl acetate group were significantly lower than that of model group(all P<0.05),LG content in the liver tissue of mice in water extract group,70%ethanol group,n-butanol group,n-butanol group,ethyl acetate group was significantly higher than that in model group(all P<0.05),the survival time of hypoxic-resistant mic
