结直肠腺癌中MMP-2和TIMP-4的表达及其意义

摘　　要：目的探讨MMP-2和TIMP-4蛋白在结直肠腺癌中的表达及其临床病理意义。方法利用qRT-PCR法和Western blot法检测20例手术切除的新鲜结直肠腺癌及其对应正常肠组织中MMP-2和TIMP-4的表达;采用免疫组化法检测90例结直肠腺癌组织中MMP-2、TIMP-4蛋白的表达,并结合临床病理特征进行分析。结果qRT-PCR实验显示MMP-2 mRNA和TIMP-4 mRNA在结直肠腺癌和正常肠组织中的表达差异有统计学意义(P<0.05)。Western blot结果显示两种蛋白在结直肠腺癌组织中高表达,在正常肠组织中低表达。免疫组化检测MMP-2和TIMP-4蛋白在结直肠腺癌、高级别上皮内瘤变、低级别上皮内瘤变和正常肠组织中的阳性率依次为81.11%和87.78%、63.33%和73.33%、40.00%和40.00%、18.00%和26.00%,两种蛋白在不同肠组织中的表达差异均有统计学意义(P<0.0001)。MMP-2蛋白表达与肿瘤大小、淋巴结转移、分化程度、TNM分期显著相关(P<0.05);TIMP-4蛋白表达与肿瘤大小、分化程度、TNM分期显著相关(P<0.05)。结直肠腺癌中MMP-2与TIMP-4蛋白表达有明显相关性(r=0.253,P=0.016)。结论MMP-2和TIMP-4在结直肠腺癌中高表达,其代谢失调可能共同参与结直肠腺癌的发生、发展过程。Purpose To investigate the expression of matrix metalloproteinase-2(MMP-2)and tissue inhibitor of metalloproteinase-4(TIMP-4)proteins in colorectal adenocarcinoma and their pathological significance.Methods The expression of MMP-2 and TIMP-4 in 20 surgically resected fresh colorectal adenocarcinomas and their paired normal tissues was detected by qRT-PCR and Western blot,respectively.The expression of MMP-2 and TIMP-4 protein was detected by immunohistochemistry in different intestinal tissues,analyzed on their clinicopathological characteristics.Results The qRT-PCR experiment showed a statistical gap in the expression of the MMP-2 mRNA and TIMP-4 mRNA in colorectal adenocarcinoma and normal intestinal tissues(P<0.05).In the Western blot experiment,the two proteins were highly expressed in colorectal adenocarcinoma tissues and lowly expressed in normal intestinal tissues.In immunohistochemistry experiments,the positive expression rates of MMP-2 and TIMP-4 protein in colorectal adenocarcinoma,high-grade intraepithelial neoplasia,low-grade intraepithelial neoplasia and normal intestinal tissue were 81.11%and 87.78%,63.33%and 73.33%,40.00%and 40.00%,18.00%and 26.00%,the differences in the expression of the two proteins in different groups were statistically significant(P<0.0001).There were statistically significant differences in the expression of the two proteins between different groups in immunohistochemical experiment(P<0.0001).MMP-2 protein expression was significantly related to tumor size,lymph node metastasis,differentiation degree,and TNM stage(P<0.05).TIMP-4 protein expression was significantly related to tumor size,differentiation degree,and TNM stage(P<0.05).There was a significant correlation between MMP-2 and TIMP-4 protein expression in colorectal adenocarcinoma(r=0.253,P=0.016).Conclusion MMP-2 and TIMP-4 are over-expressed in colorectal adenocarcinoma,and their metabolism disorders may participate in the occurrence and process of colorectal adenocarcinoma.
