摘 要:目的观察金钗石斛多糖(DNP)的对皮肤腺癌移植瘤的抗肿瘤作用并探讨其可能的抑瘤机制。方法将皮肤腺癌瘤悬液接种于BALB/c小鼠皮下,建立移植瘤模型,小鼠被随机分为4组:模型组(Model)、紫杉醇(PTX,30 mg/kg)阳性对照组、DNP低剂量(Low-DNP,50 mg/kg)和高剂量(High-DNP,200 mg/kg)组。于建模后第2天开始灌胃给药,每3天记录小鼠体质量和瘤体积,30 d后取小鼠肿瘤及各器官并计算相应指数。观察肿瘤质体及病理变化。采用RT-PCR技术检测相关凋亡基因Bax和Caspase-3来研究其可能的抗肿瘤机制。结果与Model组相比,PTX组小鼠体质量明显下降(P<0.05),而Low-DNP组和High-DNP组小鼠体质量均没有明显变化(P>0.05)。与Model组相比,PTX组显著抑制肿瘤生长(抑瘤率分别达98%)。Low-DNP组和High-DNP组均有一定的抑制肿瘤生长的作用,其抑瘤率约为50%(P<0.05)。DNP组基因Bax和Caspase-3的RNA表达量升高。组织病理学结果显示:各给药组肿瘤组织中可见不同程度的坏死,High-DNP组比Low-DNP组坏死区明显。结论DNP对皮肤腺癌荷瘤鼠有一定的抑瘤作用,其机制可能与促进肿瘤细胞凋亡有关,值得进一步研究。Objective To examine the anti-tumor effect of Dendrobium nobile polysaccharides in tumor-bearing mice.Methods Female BALB/c mice were inoculated tumor suspension subcutaneously to establish tumor-transplant model.Tumor-bearing mice were divided into four groups randomly:The model group,the positive control group treated with Paclitaxel(PTX,30 mg/kg),the low(Low-DNP,50 mg/kg)and high(High-DNP,200 mg/kg)dose of Dendrobium nobile polysaccharides,by gavages daily for 30 days.The body weight and the tumor volume were measured every 3 days.Thirty days after treatments,mice were killed and tumors and major organs were collected.Total RNA was extracted for real-time RT-PCR analysis.Results Compared with the model group,PTX almost inhibited tumor growth(98%inhibition)but also decreased animal body weights.DNP produced moderate tumor growth inhibition(about 50%inhibition),without apparent effects on animal body weight.Compared with the model group,the expressions of Caspase-3,Bax mRNA were increased by the low dose of DNP(P<0.05).Conclusion The Dendrobium nobile polysaccharides has moderate anti-tumor growth effect in skin adenocarcinoma-bearing mice,We speculate that the mechanism of DNP against transplanted skin tumor may be related to increase tumor cell apoptosis.The inhibition mechanism warrants further investigation.
