摘 要:同源染色体联会与重组是减数分裂过程中的重要生理事件,也是遗传学教学的重要知识点。在实验教学中,以小鼠睾丸为实验材料,制备减数分裂染色体展片。利用免疫荧光双标技术对联会复合体蛋白SCP3和SCP1进行标记,在荧光显微镜下观察减数分裂前期Ⅰ各阶段联会复合体的形态特点。通过对联会复合体蛋白SCP3和MLH1进行免疫荧光标记可观察到减数分裂前期Ⅰ粗线期的联会复合体和重组节。该实验项目的开设,可以帮助学生更加直观地理解同源染色体联会与DNA同源重组的概念和意义,为其将来从事雄性不育机制的科学研究和临床诊断进行一定的知识储备。通过该实验,将免疫荧光双标技术引入实验教学,还可增强学生的科研兴趣,提高课程的教学质量,为创新性人才培养模式探索提供了思路。The homologous chromosome synapsis and recombination are important physiological events in meiosis, and they are also important teaching points in genetics. In the experimental teaching, we use the testis of mouse to make slides of chromosome in meiosis firstly, and then, label the SCP3 and SCP1 by the double-immunofluorescence labeling technique to observe the morphological characteristics of synaptonemal complex in different periods of meiotic prophase Ⅰ by the fluorescence microscope. With the same method, synaptonemal complex and recombination nodule can be observed in the pachytene of meiotic prophase Ⅰ. This teaching experiment can help students to understand the concept and significance of homologous chromosome synapsis and recombination more intuitively and accumulate more knowledge for their future scientific research and clinical diagnosis of male infertility mechanism. Through this experiment, the double-immunofluorescence labeling technology is introduced to experimental teaching, and it can also enhance students’ interest in scientific research, improve the teaching quality of the course, and provide new ideas for the exploration of the innovative talent training mode.
