摘 要:利用SSR标记和形态学标记对27个叶用莴苣品种进行DNA指纹图谱分析和遗传多样性分析。从50对引物中筛选出22对引物对叶用莴苣资源进行指纹图谱构建和遗传多样性分析。形态学鉴定主要调查了26个主要的指标,主要包括叶片性状和植株性状。变性聚丙烯酰胺和毛细管电泳相结合的结果显示共扩增出187种多态性基因型,平均每对引物扩增出8.31种。引物多态信息量(PIC)分布范围为0.42~0.92,平均为0.75。用形态标记和分子标记对27个品种进行聚类分析的结果表明,分子标记聚类分析的结果均可将27份材料分为4组,而形态标记可将材料分为2组,本研究结果为叶用莴苣的品种鉴定提供了科学依据。The DNA fingerprint and genetic diversity of 27 varieties of leafy lettuce were performed by using SSR and morphological markers.From 50 pairs of primers,22 pairs of primers were selected for fingerprinting construction and genetic diversity analysis of lettuce accessions.Morphological markers included 26 indicators,which included leaf and plant traits.The integrated analysis of poly acrylamide and STR showed that 187 polymorphic bands were amplified with an average value of 8.31 bands per pair of primers.Primer polymorphism information(PIC)distribution range was 0.42~0.92,and the average value was 0.75.27 lettuce varieties could be classified to 4 groups based on morphological and 2 groups based on molecular markers.This study provides a scientific base of identification of leafy lettuce varieties.
