摘 要:拟用氨基酸、无机盐、维生素等物质替代蛋白胨,研制可质控的纯化学合成缓冲蛋白胨水培养基(BPW)。利用23种氨基酸,18种维生素,5种无机盐对BPW进行解析替代。以葡萄糖盐溶液作为基底物,在此基础上添加46种物质筛选对鼠伤寒沙门氏菌有增菌效果的物质,制成纯化学合成缓冲蛋白胨水培养基。筛选出50 mg/L硫酸镁、95 mg/L甲硫氨酸、15 mg/L半胱氨酸、32.5 mg/L胱氨酸二盐酸盐、4 g/L维生素C、10 mg/L硫酸腺嘌呤、5 mg/L硫酸锰、20 mg/L黄嘌呤8种物质,这8种物质均可以促进鼠伤寒沙门氏菌ATCC14028生长,其中硫酸镁效果最佳,OD600值达0.35;半胱氨酸、胱氨酸二盐酸盐、硫酸腺嘌呤、硫酸锰作用次之,OD600值达0.18~0.20;甲硫氨酸、维生素C作用较弱,仅达到0.15;多次筛选试验表明低浓度硫酸镁(50 mg/L)单独作用时效果最佳,故将葡萄糖盐溶液中加入低浓度硫酸镁(50 mg/L)制成纯化学合成缓冲蛋白胨水培养基。通过研究,以期为BPW培养基的生产与检定提供参考价值。In this study,a pure chemical synthesis buffer peptone medium(BPW)was developed by using amino acid,inorganic salt and vitamin instead of peptone.23 amino acids,18 vitamins,and 5 inorganic salts were used to analyze and replace the BPW medium.On the basis of salt solution and glucose,46 kinds of substances were added to screen the substances which have the effect of increasing Salmonella.These effective substances were synthesized into Buffer Peptone Water pure chemical defined medium.50 mg/L magnesium sulfate,95 mg/L methionine,15 mg/L cysteine,32.5 mg/L cystine dihydrochloride,4 g/L vitamin C,10 mg/L Adenine Sulfate,5 mg/L manganese sulfate and 20 mg/L xanthine could promote the growth of S.typhimurium ATCC14028.Magnesium sulfate had the best effect,OD600 value was 0.35;cysteine,cystine dihydrochloride,Adenine Sulfate and manganese sulfate had the second effect,OD600 value was 0.18-0.2;Methionine and vitamin C had a weak effect,OD600 value only reached 0.15.Several screening experiments showed that the effect of magnesium sulfate(50 mg/L)with low concentration was the best.Therefore,glucose salt solution was added with magnesium sulfate to make a purely chemical defined medium.Glucose salt solution was added with low-concentration magnesium sulfate(50 mg/L)as a pure chemical defined medium with quality control Buffered Peptone Water medium.This study provides reference value for the production and verification of BPWmedium.
